Expressing nktGD8239 under the control of either Scer\GAL4repo.PU, Scer\GAL4elav.PU or Scer\GAL4nSyb.PU results in a significant decrease in night-time sleep, with a reciprocal increase in night-time locomotor activity, but no effect on day-time sleep; there is a significant increase in sleep latency and fragmented sleep, with significantly more but shorter sleep bouts. In the Scer\GAL4repo.PU and Scer\GAL4elav.PU conditions there is normal circadian rhythmicity (i.e. it is maintained under constant darkness) and normal waking activity index. In the Scer\GAL4elav.PU and Scer\GAL4nSyb.PU conditions there is normal sleep loss and gain after 12 h of sleep deprivation. Introducing Scer\GAL80Mef2.mb247 rescues the sleep phenotype observed upon Scer\GAL4nSyb.PU-driven nktGD8239 expression.

Expressing nktGD8239 under the control of Scer\GAL4GMR86E01, Scer\GAL4238Y, Scer\GAL4c305a or Scer\GAL4GMR35B12 leads to a decrease in night-time sleep, but no effect on day-time sleep.

Adulthood-only expression of nktGD8239 under the control of either Scer\GAL4repo.PU or Scer\GAL4nSyb.PU (temporally regulated by Gal80[ts] and temperature shift from 18[o]C to 30[o]C) leads to a decrease in night-time sleep, but no effect on day-time sleep. The night-time sleep is not severely fragmented: in the Scer\GAL4nSyb.PU condition there is no effect in sleep bout number or length, while in the Scer\GAL4nSyb.PU condition there is a decrease in sleep bout length, but not number.

Expressing nktGD8239 under the control of Scer\GAL4NP3233 alone, Scer\GAL4Tub.PU alone or Scer\GAL4Eaat1.PR and Scer\GAL80elav.PU combined leads to a significant increase in locomotor activity during night-time; this is not observed if nktGD8239 is expressed under the control of Scer\GAL4NP2222 alone or Scer\GAL4Mz709 alone.

Adults expressing CG14141GD8239 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Depending on the insertion line used, expression under the control of Scer\GAL4Mef2.PR can result in viable flies or semi-lethality.
