UASt regulatory sequences drive expression of an inverted repeat.
wing, with Scer\GAL469B
Expressing Dis3l2GD9240 under the control of Scer\GAL469B leads to a large wing.
Expression of Dis3l2GD9240 under the control of any of the following: Scer\GAL469B, Scer\GAL4nub-AC-62 or Scer\GAL4da.G32 results in a significant increase of adult wing size, expression driven by Scer\GAL4en.PU leads to enlarged posterior wing compartment but to a lesser extent also the anterior compartment. Expression under either Scer\GAL4Act5C.PI or Scer\GAL4αTub84B.PL also increases the adult wing size even though the overall body mass is not different from the controls (with Scer\GAL4Act5C.PI). Expression of Dis3l2GD9240 (with Scer\GAL469B or Scer\GAL4nub-AC-62) increases the wing cell number whereas the cell density is not changed relative to controls. Expression with the Scer\GAL469B driver combined with Gal80[ts] to exert temporal control during second and early third larval instar leads to increased wing size in the adults, but from mid third larval instar onward the Dis3l2GD9240 expression fails to induce wing overgrowth. Larval wing disc expressing Dis3l2GD9240 (either with the Scer\GAL469B driver or with Scer\GAL4nub-AC-62) are enlarged compared to controls due to increase in cell proliferation (assessed by pH3 staining)
Adults expressing CG16940GD9240 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.
Dis3l2GD9240, Scer\GAL469B has hyperplasia | adult stage phenotype, enhanceable by Df(3L)H99/+
Scer\GAL469B/Dis3l2GD9240 is a suppressor of abnormal size | adult stage phenotype of pcm5
Scer\GAL469B/Dis3l2GD9240 is a suppressor of abnormal size | third instar larval stage phenotype of pcm14
Scer\GAL469B/Dis3l2GD9240 is a non-suppressor of abnormal developmental rate | larval stage phenotype of pcm14
Scer\GAL469B/Dis3l2GD9240 is a suppressor of wing phenotype of pcm5
Scer\GAL469B/Dis3l2GD9240 is a suppressor of wing disc | third instar larval stage phenotype of pcm14
The increased adult wing size observed in both Df(3L)H99 heterozygotes and flies expressing Dis3l2GD9240 driven by Scer\GAL469B is increased still further if the manipulations are combined.
The reduced wing size observed in pcm5 mutant adults is fully rescued by expression of Dis3l2GD9240 under the control of Scer\GAL469B in the mutant background. Similarly, the small wing discs size characteristic for pcm14 mutant third instar larvae is also suppressed by expression of Dis3l2GD9240 but the larvae are still developmentally delayed.
