Expression of SypGD9477 under the control of Scer\GAL4GMR.PU has no obvious effect on adult eye morphology, whereas expression driven by Scer\GAL4elav.PU results in complete paralysis of adult flies.

Expression of SypGD9477 driven by Scer\GAL4RapGAP1-OK6 does not significantly affect bouton number (or type Is or Ib bouton numbers) at the neuromuscular junction (NMJ) of third instar larvae, compared to controls. When expression of SypGD9477 is driven in muscle by Scer\GAL4Mhc.PW, there are significant increases in total bouton number (and type Is but not Ib boutons) compared to controls. When expression of SypGD9477 is driven pan-neuronally by Scer\GAL4elav-C155, there is significant NMJ overgrowth (increases in branch and bouton numbers) compared to controls.

Expression under the control of Scer\GAL41407 results in shorter neuroblast lineages (less daughter cells per neuroblast) in the larval brain compared to controls.

Adults expressing SypGD9477 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Expression of SypGD9477 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) results in viable flies or complete lethality, depending on the particular P{GD9477} insertion line used.

Depending on the insertion line used, expression under the control of Scer\GAL4Mef2.PR can result in early pupal lethality or late larval lethality.

Expression under the control of Scer\GAL4Mef2.PR results in grossly normal larval body wall muscles.

Expression under the control of Scer\GAL4Mef2.PR results in sarcomeres with reduced Z-lines in the larval body wall muscles.

Expression under the control of Scer\GAL4pnr-MD237 results in bristle morphology defects on the notum in 0% or 20-30% of the Scer\GAL4pnr-MD237 expression domain, depending on the insertion line used.

Expression under the control of Scer\GAL4pnr-MD237 results in the absence of 80-90% or 90-100% of the Scer\GAL4pnr-MD237-expressing area of the notum, depending on the insertion line used.

Adult flies expressing SypGD9477 (together with UAS-Dicer2 to enhance RNAi efficiency) under the control of Scer\GAL4elav.PU in either wild-type or TBPHΔ23/+; TBPHdsRNA.Scer\UAS.cUa background appear to be completely paralyzed.

The eye degeneration phenotype (appearance of melanotic spots, fusion of ommatidia) characteristic for adult flies expressing TBPHScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4GMR.PU is strongly suppressed by co-expression of SypGD9477.
