Reference

Expression of Nep1GD11797 under the control of either Scer\GAL4c739 or Scer\GAL4VT064246 (using tub-Gal80[ts] to limit the expression to adulthood) results in significantly reduced middle-term memory compared to controls (tested in an aversive olfactory memory assay 2hr post-training). Scer\GAL4VT064246-driven expression also decreases the long-term (24hr) memory.

Wild type females mated to males expressing Nep1GD11797 via Scer\GAL4tub.PU or Scer\GAL4hs.2sev, using Scer\GAL80ts.αTub84B to suppress Scer\GAL4 function during development, lay significantly fewer eggs than controls. However, no effect on the proportion of progeny that eclose from laid eggs (hatchability) is observed.

Wild type males mated to females expressing Nep1GD11797 via Scer\GAL4tub.PU or Scer\GAL4hs.2sev, using Scer\GAL80ts.αTub84B to suppress Scer\GAL4 function during development, lay significantly fewer eggs than controls. No effect on the proportion of overall progeny that eclose from laid eggs (hatchability) is observed, though early hatchability is reduced - the severity is dependent on the insertion used.

Wild type males mated to females expressing Nep1GD11797 via Scer\GAL4nSyb.PS or Scer\GAL4Send1.4kb, using Scer\GAL80ts.αTub84B to suppress Scer\GAL4 function during development, lay fewer eggs than controls. However, the proportion of progeny that eclose from laid eggs (hatchability) is not affected.

Adults expressing Nep1GD11797 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Depending on the insertion line used, expression under the control of Scer\GAL4Mef2.PR can result in viable flies or late pupal lethality.

The impaired middle- or long-term term memory, MTM or LTM, tested in an aversive olfactory memory assay 2hr or 24hr post-training, respectively, characteristic for adult flies expressing Nep1GD11797 under the control of either Scer\GAL4c739 or Scer\GAL4VT064246 (combined with tub-Gal80[ts] to limit expression to adulthood) is not by enhanced by co-expression of ApplAβ.Scer\UAS.T:Ivir\HA1 with the Scer\GAL4c739 driver, but significantly worsens the MTM phenotype with the Scer\GAL4VT064246 driver. Flies simultaneously expressing Nep1GD11797 and ApplAβ.Scer\UAS.T:Ivir\HA1 driven by Scer\GAL4VT064246 do not show any learning ability defects. Both the MTM and the LTM deficit observed in flies expressing Nep1GD11797 under Scer\GAL4VT064246 are exacerbated further by co-expression of BaceScer\UAS.cCSa.
