Statement Reference Knockdown of Pdf through expression of Pdf[GD299] under the control of Scer\GAL4[tim.Scer\UAS] results in a clearly advanced phase of the evening activity peak in LD 12:12. Expression under the control of Scer\GAL4[pnr-MD237] may result in semi-lethality, depending on the insertion line used.
Expression under the control of Scer\GAL4[pnr-MD237] results in a colour difference between the central Scer\GAL4[pnr-MD237] expression domain of the notum and the surrounding lateral region in 0% or 60-70% of the Scer\GAL4[pnr-MD237] expression domain, depending on the insertion line used. | |||

Statement Reference In LD 12:12, the evening activity peak of npf[KK111846]/Pdf[GD299] knockdown flies (under the control of Scer\GAL4[tim.Scer\UAS]) is different from Pdf[GD299]-knockdown flies: it appears broader and flatter. Compared to control lines, the Dcr-2[Scer\UAS.cDa]/Scer\GAL4[tim.Scer\UAS]/Pdf[GD299] flies show increased arhythmicity and shortened free-running period under constant darkness. Under light/dark cycles, these flies display reduced morning anticipation and an increase in activity levels in the six hours before lights-off, which results in an advanced evening peak.
Dcr-2[Scer\UAS.cDa]/Scer\GAL4[R6]/Pdf[GD299] flies display high levels of arhythmicity under constant darkness. Under light/dark cycles, these flies display significantly reduced morning anticipation controls lacking Scer\GAL4[R6] or Pdf[GD299].
Dcr-2[Scer\UAS.cDa]/Scer\GAL4[Mai179]/Pdf[GD299] flies display high levels of arhythmicity under constant darkness. These flies show a normal free-running period under constant darkness and normally phased evening peak under light/dark cycles. | |||
