The expression of UchGD11265 under the control of Scer\GAL4ple.PF results in locomotor dysfunction and deficit in both larvae and adults, loss of larval DL1 cluster DA neurons, but not DM1a, DM1b, DM2, DL2a or DL2b cluster DA neurons, and loss of adult PPM2, PPM3 (only from the PPM3b sub-cluster), PPL2ab and VUM cluster DA neurons, as compared to controls. The expression of UchGD11265 also results in pupal lethality (driven by Scer\GAL4Act5C.PI), in a rough eye phenotype (driven by Scer\GAL4GMR.PS), in an abnormal vein pattern (driven by Scer\GAL4Bx-MS1096), in abnormal bristle and hair patterns (driven by Scer\GAL4pnr-MD237), in a progressive locomotor deficit (driven by Scer\GAL4Ddc.PL), and in an abnormal morphology larval neuromuscular junction morphology, namely longer branches and fewer and bigger type Ib boutons (driven by Scer\GAL4Toll-6-D42), as compared to controls.
No phenotype is observed when UchGD11265 is expressed using Scer\GAL4sqh.PW or Scer\GAL4elav.PU.
Adults expressing UchGD11265 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.
