Expression of Xbp1GD4745 under the control of Scer\GAL4Su(H).GBE (but not under either Scer\GAL4ppl.PU or Scer\GAL4how-24B) and using tub-Gal80[ts] to limit the time of expression, increases cell proliferation in the adult gut.

The temporally regulated expression (by Gal80[ts]) of Xbp1GD4745, for 5 days under the control of Scer\GAL4NP0001, or for 6 days under the control of Scer\GAL4NP5130, under the combined control of Scer\GAL4NP5130 and Scer\GAL80Su(H).GBE or under the control of Scer\GAL4Su(H).GBE, results in a strong increase in the proportion of mitotic cells in adult midguts, as compared to controls; expression for 6 days under the control of Scer\GAL4NP5130 also results in an increase in the population of intestinal stem cells, assessed by immmuno-staining for Delta and esg, in adult midguts as compared to controls. Clonal expression in the adult midgut leads to a strong increase in the average clone cell number and in the proportion of mitotic cell per midgut at 6 days after clone induction (when expression is under the control of Scer\GAL4Act.PU) and leads to a strong increase in the proportion of mitotic cell per midgut at 4 and 7 days after clone induction (when expression is under the control of Scer\GAL4unspecified), as compared to control clones.

Flies expressing Xbp1GD4745 in the developing eye, under the control of Scer\GAL4GMR.PF, exhibit slightly irregular and depigmented eyes but display mainly normal retinae.

Expression of Xbp1GD4745 using Scer\GAL4ap-md544 results in semi-lethality. Adult survivors have severely disrupted wings which appear as small amorphic inflated structures, accompanied by marked excess of bristles on the wing and scutum, while the scutellum is not affected.

Expression of Xbp1GD4745 using Scer\GAL4pnr-MD237 results in excess bristles on the mid-line of the scutum while the wings are not affected. No extra bristles are observed on the scutellum; some flies even exhibit a reduced number of scutellar bristles.

Expression of Xbp1GD4745 under the control of Scer\GAL4elav-C155 slightly increases the level of apoptosis in the developing nervous system.

Adults expressing Xbp1GD4745 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Expression under the control of Scer\GAL4pnr-MD237 results in extra bristles on the notum in 80-90% of the Scer\GAL4pnr-MD237 expression domain.

Expression under the control of Scer\GAL4pnr-MD237 results in external sensory organs consisting of empty or multiple sockets in 30-40% of the Scer\GAL4pnr-MD237 expression domain of the notum.
