Scer\GAL4Mef2.PU-mediated expression of parkGD5543 results in the appearance of swollen/clumped mitochondria in adult indirect flight muscles. EM analysis reveals vacuolization of adult flight muscle cells and severely swollen mitochondria with disrupted cristae. Mitochondrial membrane potential in Scer\GAL4tub.PU parkGD5543 flies is reduced, and 2-week-old adult flies show a defect in climbing ability.

Adults expressing parkGD5543 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Expression of parkGD5543 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) results in viable flies or partial lethality, depending on the particular P{GD5543} insertion line used.

Depending on the insertion line used, expression under the control of Scer\GAL4Mef2.PR can result in flightless flies.

Expression under the control of Scer\GAL4Mef2.PR results in missing adult indirect flight muscles.
