Expression of sbbGD10734 under the control of Scer\GAL4N630 (in combination with a heat shock to induce expression) leads to a decrease in the rate of male copulation success, and a wing vein phenotype wherein wing vein L5 fails to reach the wing margin.

Expression of sbbGD10734 under the control of Scer\GAL4l.Trh.PK in males leads to loss of serotonergic dsx-positive neurons in the abdominal ganglion, as compared to controls.

Expression of sbbGD10734 under the control of Scer\GAL4fru.P1.D or Scer\GAL4fru-NP0021 (in combination with a Dicer-2 transgene to enhance RNAi efficiency) leads to a significant decrease in the rate of male copulation success, and expression under the control of Scer\GAL4fru.P1.D leads to a decrease in courtship index, however, expression of sbbGD10734 under the control of Scer\GAL4fru-NP0021 does not lead to any significant decrease in courtship index. Expression of sbbGD10734 under the combinatorial control of Scer\GAL4fru.P1.D, Scer\GAL80Scer\FRT.αTub84B and Scer\FLP1dsx does not lead to male courtship or copulation defects.

Expression of sbbGD10734 under the control of Scer\GAL4dsx.KI (with or without a Dicer-2 transgene to enhance RNAi efficiency) leads to a total lack of male copulation success and a significant decrease in courtship index, as compared to controls, but this phenotype is not seen upon inclusion of a Scer\GAL80elav.PS transgene. Expression of sbbGD10734 under the combinatorial control of Scer\GAL4dsx.KI and Scer\GAL80repo.PL, or of Scer\GAL4dsx.KI, Scer\GAL80Scer\FRT.αTub84B and Scer\FLP1o.FLP.oc.T:SV40\nls-largeT, results in a complete lack of male copulation success, but no significant decrease in courtship index, as compared to controls. Expression of sbbGD10734 under the combinatorial control of Scer\GAL4dsx.KI, Scer\GAL80αTub84B.PG and Scer\FLP1fru, or of Scer\GAL4dsx.KI, Scer\FLP1o.FLP.oc.T:SV40\nls-largeT and Scer\GAL80αTub84B.PG, does not lead to male courtship or copulation defects. Expression of sbbGD10734 under the combinatorial control of Scer\GAL4dsx.KI, Scer\GAL80IVS.WPRE.8xEcol\lexAop, and Ecol\lexAfru.P1.T:Hsim\VP16, or of Scer\GAL4dsx.KI and Scer\GAL80tsh-GAL80, results in a complete lack of male copulation success, and a decrease in courtship index, as compared to controls.

Expression of sbbGD10734 under the combinatorial control of Scer\GAL80αTub84B.PG and Scer\FLP1dsx with Scer\GAL4Poxn.14, Scer\GAL4VGlut-OK371, Scer\GAL4VGlut.PD, Scer\GAL4ChAT.7.4, Scer\GAL4ple.PF, Scer\GAL4l.Trh.PK, Scer\GAL4Gad1.3.098 or Scer\GAL4Gad1.PS (but not with Scer\GAL4ppk.PU or Scer\GAL4Tdc2.PC) results in a complete lack of male copulation success. Expression of sbbGD10734 under the combinatorial control of Scer\GAL80αTub84B.PG and Scer\FLP1dsx with either Scer\GAL4Poxn.14 or Scer\GAL4ppk.PU does not cause a significant decrease in courtship index, as compared to controls.

Adults expressing sbbGD10734 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) show significantly reduced avoidance of noxious temperature (46[o]C) compared to control flies.
