Amino acid replacement: D945A.

UASt regulatory sequences drive expression of a mutated form of Gyc76C. The Gyc76C sequence has a D945A mutation and also has two Tag:MYC epitopes in place of the N-terminal signal sequence. The amino acid change is designed to disrupt the Gyc76C product's cyclase activity.

Expression of Gyc76CD945A.UAS.Tag:MYC under the control of Scer\GAL4Cg.PA leads to an increase in the number of proliferating hemocytes in third instar larval hemolymph when compared to controls.

Expression of Gyc76CD945A.Scer\UAS.T:Hsap\MYC under the control of either the Scer\GAL4A9 or the Scer\GAL4hh.PU driver does not induce ectopic venation but on the contrary results in partial or complete loss of posterior crossvein in adult wings.

Expression of Gyc76CD945A.Scer\UAS.T:Hsap\MYC in all neurons, driven by Scer\GAL4elav.PLu, perturbs motor and CNS axon guidance in embryos. For example, axons of the ISNb and SNa fail to reach their proper targets and the RP3 axon extends exuberantly in the cleft between muscles 6 and 7.
