Expression of ds[ecto.Scer\UAS] under the control of Scer\GAL4[ptc-559.1] and Scer\GAL80[ts.αTub84B] at 17[o]C has no effect on denticle belt polarity. However, growth at 29[o]C results in severe denticle polarity defects. Defects are also seen when the temperature is shifted from 17[o]C to 29[o]C before the moult to second stage larva. The converse shift, from 29[o]C to 17[o]C at the second instar larval stage results in wild-type denticle belt polarity.
Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) that are expressing dsecto.Scer\UAS under the control of Scer\GAL4αTub84B.PL in a wild-type background usually result in a reversal of cell polarity in wild-type cells posterior to the clone (hairs behind the clone point forwards), with a range of several cells. Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) that are expressing dsecto.Scer\UAS under the control of Scer\GAL4αTub84B.PL in a dsunspecified background do not change the cell polarity of cells surrounding the clone. Clones in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are mutant for dsunspecified and are also expressing dsecto.Scer\UAS under the control of Scer\GAL4αTub84B.PL can reverse the cell polarity of cells posterior to the clone.

Reference
ds[UAO71] ft[15] double mutant clones in the dorsal adult abdomen where the clones are also expressing ds[ecto.Scer\UAS] under the control of Scer\GAL4[αTub84B.PL] strongly repolarise hairs behind the clone, reversing their polarity (100% of clones repolarise with a maximum range of up to seven cells). This repolarisation is largely suppressed by co-expression of fj[Scer\UAS.cZa] within the clone (48% of clones show some repolarisation with a range of one to three cells).
The reversal of cell polarity that is seen in cells posterior to clones in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are expressing dsecto.Scer\UAS under the control of Scer\GAL4αTub84B.PL is still seen if the clones are induced in a stan- background (the cells within the clone are stanE59/stanE59 while the cells surrounding the clone are stan3/stanE59). The reversal of cell polarity that is seen in cells posterior to clones in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are expressing dsecto.Scer\UAS under the control of Scer\GAL4αTub84B.PL is still seen if the clones are induced in a fzunspecified background. The reversal of cell polarity that is seen in cells posterior to clones in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are expressing dsecto.Scer\UAS under the control of Scer\GAL4αTub84B.PL is still seen if the clones are induced in a fzunspecified stan- background (cells within the clone are stanE59/stanE59 while the cells surrounding the clone are stan3/stanE59). Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) that are expressing dsecto.Scer\UAS under the control of Scer\GAL4αTub84B.PL in a ft12/ftunspecified background do not change the cell polarity of cells surrounding the clone. Clones in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are mutant for ftunspecified and are also expressing dsecto.Scer\UAS under the control of Scer\GAL4αTub84B.PL can reverse the cell polarity of cells posterior to the clone. Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are co-expressing both fjScer\UAS.cZa and dsecto.Scer\UAS under the control of Scer\GAL4αTub84B.PL result in a reversal of cell polarity in cells anterior to the clone. Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are co-expressing both ftScer\UAS.cMa and dsecto.Scer\UAS under the control of Scer\GAL4αTub84B.PL result in a reversal of cell polarity in cells anterior to the clone. The reversal of cell polarity that is seen in cells posterior to clones in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are expressing dsecto.Scer\UAS under the control of Scer\GAL4αTub84B.PL is enhanced if the clones are induced in a fjunspecified background compared to a wild-type background; the repolarisation occurs over a greater range in the fjunspecified background.
