Drives expression in alpha' and beta' lobes of the mushroom body, and in alpha'/beta' mushroom body neurons.

ScerGAL4c305a labels a large cluster of excitatory local interneurons with cell bodies located lateral to the antennal lobe. Their axons fasciculate and enter the antennal lobe.

Expression of ScerGAL4c305a was observed in the alpha' and beta' lobes of the mushroom body at 4 days after larval hatching (late wandering third instar), but not earlier at 2 days after hatching (early third instar).

ScerGAL4c305a labels the mushroom body alpha'/beta' neurons. Co-expression with a postsynaptic marker labels Kenyon cell terminals in the mushroom body alpha' and beta' lobes. No presynaptic terminals in the calyx are observed.

This line labels larval-born alpha' and beta' Kenyon cells in the third instar as well as elements in the thoracic and abdominal ganglia, mostly sensory terminals from the periphery.

Expression is observed in about half of the the alpha', and beta' neurons of the adult mushroom body, in ellipsoid body neurons and in the antennal lobe.

Expression of ScerGAL4c305a in the adult antennal nerve is in putative mechanosensory neurons, and within the ring neurons of the ellipsoid body.

    Detailed Description
    Statement
    Reference

The inhibition of α'/β' class Kenyon cells by the expression of shits.UAS.cUa under the control of Scer\GAL4c305a at the restrictive temperature of 29[o]C leads to third instar foraging larvae showing increased naive chemotactic response, as well as decreased olfactory habituation, to ethyl acetate.

Inhibition (using shi1.UAS at 32[o]C) of Scer\GAL4c305a cells in adults significantly reduces startle-induced climbing behavior, compared to controls.

Thermogenetic activation (using TrpA1UAS.(B).cKa at 32[o]C) of Scer\GAL4c305a cells in adults significantly reduces startle-induced climbing behavior, compared to controls.

Inhibition (using shi1.UAS at 31[o]C) of Scer\GAL4c305a cells in adults during consolidation only, but not testing only, significantly reduces 3-hour aversive olfactory anaesthesia-resistant memory, compared to controls.

Activation of Scer\GAL4c305a-expressing mushroom body α/β neurons results in a strong decrease in nighttime sleep. This nighttime sleep loss remains stable over 48 hours of activation and is accompanied by increasing daytime sleep loss.

Inhibition (using shi1.UAS at 32[o]C) of Scer\GAL4c305a cells in adults does not affect 5 min (single-trial conditioned) or 30 min (multiple-trial conditioned) aversive taste memory, compared to controls.

Inhibition of neural activity in Scer\GAL4c305a-expressing neurons does not affect sensitivity to (S,Z,Z)-CH503 (a more potent stereoisomer of the gustatory sex pheromone CH503). As in wild type flies, courtship defects are seen in response to (S,Z,Z)-CH503.

Inhibition (using shi1.UAS at 33[o]C) of Scer\GAL4c305a cells in adults significantly reduces aversive 1-min olfactory, but not visual, memory, compared to controls.

Inhibition (using shi1.UAS at 33[o]C) of Scer\GAL4c305a cells in adults during testing does not affect 3 h aversive or appetitive memory, compared to controls.

Inhibition (using shi1.UAS at 31[o]C) of Scer\GAL4c305a cells in adults does not affect short-term aversive olfactory or visual unimodal memory, compared to controls; inhibition does not affect short-term aversive memory for tested olfactory cues that are associated with conditioned visual cues, compared to controls.
