ScerGAL4ppk.PG drives expression strongly in class IV dendritic arborization neurons and weakly in class III neurons.

ScerGAL4ppk.PG drives expression in larval multidendritic neurons (Type II). All of the Type II neurons identified by ScerGAL4ppk.PG are also labeled with ScerGAL4Piezo.1.0.

ScerGAL4ppk.PG drives expression in all class IV dendritic arborizing neurons which co-express NcraQFTrpA1.PP.

ScerGAL4ppk.PG drives expression in peripheral sensory neurons in the legs, wings, body wall, and reproductive tract. In the reproductive tract it is expressed in 2-3 fruitless expressing neurons on either side of the uterus as well as a small number of neurons surrounding the oviduct. The reproductive tract neurons project fine processes between the muscle and epithelial layers that enter and arborize within the lumen of the uterus while other processes bifurcate close to the soma and innervate the lower regions of the common oviduct, both these aborizations run along the inner surface of the epithelial cell layer. Infrequently, expression is also seen in 1-2 neurons near the base of the antennal nerve and the the lateral horn. Afferents from all of the above neurons contribute to the abdominal nerve trunk, most terminating in the thoracico-abdominal ganglion but some extend further in an anterior direction, and a pair of bilateral fibers emerge from the network in the abdominal ganglion, traverse the entire length of the ventral side of the nerve cord, extend through the cervical connective before terminating in the posterior region of the adult gnathal ganglion.

ScerGAL4ppk.PG drives expression in the adult peripheral neurons, including 2 neurons on each lateral oviduct and 2-3 neurons on each side of the uterus, organised in 3 clusters.

Inhibiting the activity of ppk neurons (by the constitutive or clonal expression of Ctet\tetXTNT-LC.UAS.cSa under the control of Scer\GAL4ppk.PG) leads to females being less receptive to mating than controls, while still being receptive to mate (and with increased egg retention) after injection with sex-peptide.

Optogenetic inhibition of larval (Scer\GAL4ppk.PG-positive) nociceptors, by expressing Gthe\ACR1UAS.d.EYFP under the control of Scer\GAL4ppk.PG, leads to reduced nociceptive behavior to AITC (decreased larval rolling).

Inhibition (using Hsap\KCNJ2UAS.EGFP) of Scer\GAL4ppk.PG-expressing neurons respectively abolishes and severely decreases larval responses (rolling) to harsh touch (44mN) and noxious heat (39.5[o]C).

Hyperpolarization (using Hsap\KCNJ2UAS.EGFP) of Scer\GAL4ppk.PG cells in larvae significantly reduces nociceptive movement responses following stimulation with a von Frey filament, compared to controls; there is no effect on the response to gentle touch and there is a small, but significant, increase in locomotion speed, compared to controls.

Inhibition (using Ctet\tetXTNT-LC.UAS) of Scer\GAL4ppk.PG cells in larvae significantly reduces nociceptive movement responses following stimulation with a von Frey filament and increases the latency of rolling in response to high temperature (local) stimulation, compared to controls.

Optogenetic activation (using Cnoc\ChR1::Csub\ChRCsChrimson.IVS.20xUAS.Venus with 625 nm (red) light) of Scer\GAL4ppk.PG cells in larvae significantly increases rolling responses in the absence of pain stimuli, compared to controls; expression of Cnoc\ChR1::Csub\ChRCsChrimson.IVS.20xUAS.Venus in Scer\GAL4ppk.PG neurons results in their partial degeneration in the larva.

Inhibition (using shi1.UAS at 31[o]C) of Scer\GAL4ppk.PG cells, or the combined set of Scer\GAL4ppk.PG and Scer\GAL419-12 cells, in larvae significantly reduces the probability of backwards motion in response to a head touch, compared to controls.

Silencing neurons expressing Scer\GAL4ppk.PG results in reduced locomotor turning frequency and thermal nociception defects in third instar larvae.

Inhibition (using shi1.UAS at 30[o]C) of Scer\GAL4ppk.PG cells significantly reduces mating receptivity and increases egg laying in virgin females, to the extent that egg laying occurs at a similar rate to that seen in mated females, compared to controls, with no effect on egg laying in mated females.
