Constitutive overexpression of Gadd45[Scer\UAS.P\T.cPa] under the control of Scer\GAL4[1407] results in a lifespan increase in both males and females compared to controls. The median lifespan of males with constitutive Gadd45[Scer\UAS.P\T.cPa] overexpression in the nervous system is extended by 77 and 73% compared to the median lifepsan of males of the parental Gadd45[Scer\UAS.P\T.cPa] and Scer\GAL4[1407] strains, respectively. The age of 90% mortality of males with constitutive Gadd45[Scer\UAS.P\T.cPa] overexpression is increased by 25 and 59% compared to parental Gadd45[Scer\UAS.P\T.cPa] and Scer\GAL4[1407] males, and in females by 11 and 50%, respectively. This indicates that constitutive Gadd45[Scer\UAS.P\T.cPa] overexpression in the nervous system increases lifespan and delays aging in males and females. This effect is larger in males than in females. Survival curves demonstrate a lifespan increase in males expressing Gadd45[Scer\UAS.P\T.cPa] in neuronal cells under the control of Scer\GAL4[elav.Switch.PO] that are treated with mifepristone compared to those kept with mifepristone and males of parental strains. Specifically, the median lifespan of males with mifepristone-inducible Gadd45[Scer\UAS.P\T.cPa] overexpression in the nervous sytem is higher by 40-42% compared to males with the same genotype not treated with mifepristone, while in females, by 3-6%. The median lifespan of males with conditional Gadd45[Scer\UAS.P\T.cPa] overexpression in the nervous is increased by 50-70 and 62-84%, compared to the median lifespan of parental Gadd45[Scer\UAS.P\T.cPa] and Scer\GAL4[elav.Switch.PO] strain males, respectively, and in females by 27-29 and 100-102%, respectively. In females with constitutive Gadd45[Scer\UAS.P\T.cPa] overexpression in the nervous system (under the control of Scer\GAL4[1407]), fecundity is higher over the entire course of lifetime, compared to the parental strain females. The average number of eggs laid daily by a female increases 2-2.3-fold compared to parental strains, while the number of pupae per female is 3.7 and 4.4 times higher, respectively. In females with Gadd45[Scer\UAS.P\T.cPa] overexpression in the nervous system (under the control of Scer\GAL4[elav.Switch.PO]), the average number of eggs laid daily increase 1.7 times compared to the Scer\GAL4[elav.Switch.PO] parental strain. The locomotor activity of males and females with constitutive Gadd45[Scer\UAS.P\T.cPa] overexpression in the nervous system (under the control of Scer\GAL4[1407]) is increased compared with males and females of both parental strains throughout their whole lifetime, both in terms of spontaneous locomotor activity and according to a negative geotaxis test. There is an increase in locomotor activity in males (on day 4-29) and in females (on day 12-36) with conditional expression of Gadd45[Scer\UAS.P\T.cPa] under the control of Scer\GAL4[elav.Switch.PO]. In the neuroblasts of larvae constitutively or conditionally expressing Gadd45[Scer\UAS.P\T.cPa] in the nervous system under the control of Scer\GAL4[1407] or Scer\GAL4[elav.Switch.PO] respectively, the DNA damage level is reduced by 21-27%, with statistical significance compared to control larvae. However, the level of apoptosis seen in these larval neuroblasts does not differ significantly.
Expression of Gadd45[Scer\UAS.P\T.cPa] under the control of Scer\GAL4[CY2] results in apoptosis of the egg chamber, shown by condensation and breaking up of DNA within both the nurse and follicle cells. Expression of Gadd45[Scer\UAS.P\T.cPa] under the control of Scer\GAL4[GMR.PF] does not result in any gross morphological changes in the eye. Expression of Gadd45[Scer\UAS.P\T.cPa] under the control of Scer\GAL4[nos.UTR.T:Hsim\VP16] results in a broad range of abnormal eggshell phenotypes, ranging from normal morphology to dorsalised. Weakly dorsalised eggs have two broadened dorsal appendages, while moderately dorsalised eggshells have one broadened dorsal appendage on the dorsal midline. A small fraction of the eggs have a more posteriorly located patch of dorsal appendage material.
