Expression of TBPHScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4GMR.PU leads to eye degeneration in adult flies with loss of pigmentation, dark melanotic spots and ommatidial fusion.

Adult flies expressing TBPHScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4GMR.PU show large necrotic patches in the eyes.

Flies expressing TBPHScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4elav.PU, restricted to the adult stage using Scer\GAL80ts.αTub84B, exhibit serious climbing defects compared to controls.

Expression of TBPHScer\UAS.T:Zzzz\FLAG in the nervous system under the control of Scer\GAL4nSyb.PU results in temperature dependent lethality. At lower temperatures, immature-looking escapers are seen with unexpanded wings, soft cuticle, unretracted ptilinum, a dimpled dorsal thorax and misoriented scutellar bristles. Lethality at high temperatures and wing phenotypes are also seen with Scer\GAL4elav-C155 and when TBPHScer\UAS.T:Zzzz\FLAG is expressed in the motor neurons under the control of Scer\GAL4D42. Temperature-dependent lethality and a mild wing inflation phenotype is also seen when TBPHScer\UAS.T:Zzzz\FLAG is expressed in CCAP/bursicon neurons under the control of Scer\GAL4CCAP.PP.

Expression of TBPHScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4CCAP.PP has no effect on the morphology or number of CCAP neurons in third instar larvae or pupae up to 60 hours after puparium formation. Outgrowth of adult neurites is normal at 60 hours APF. However, at 72h APF and in newly eclosed adults, the number of CCAP/bursicon neurons is reduced.

Expression of Diap1Scer\UAS.cHa suppresses the reduction in CCAP/bursicon neuron number seen in newly eclosed flies expressing TBPHScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4CCAP.PP. The wing inflation defects are also rescued.

Expression of Map205GD8458 suppresses the immature wing inflation phenotype seen when TBPHScer\UAS.T:Zzzz\FLAG is expressed under the control of Scer\GAL4nSyb.PU.

Expression of Map205GD8458 suppresses the wing inflation phenotype and loss of CCAP/bursicon neurons seen when TBPHScer\UAS.T:Zzzz\FLAG is expressed under the control of Scer\GAL4CCAP.PP.

Glial expression of TBPHScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4repo.PU significantly rescued the morphological defects in glial shape, the alterations in locomotive behavior and the life span reduction associated with TBPHΔ23 from larval stages to adulthood.

Expression of TBPHScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4elav.Switch.PO (fed 0.05 or 0.25ul RU486 throughout the larval stages) is sufficient to rescue the larval locomotion, NMJ growth and cytoskeletal defects in synaptic boutons seen in TBPHΔ23 mutants. No rescue is seen when the flies are fed low doses of RU486 of ethanol. When expression is restricted to either 48 or 72 hours after egg laying the larval locomotion and NMJ assembly is not rescued at the third instar larval stage. Rescue can occur when drug treatment ends 12 hours prior to measurement. 12 hours of RU486 treatment in mature third instar larval stages until the end of the larval period is sufficient to restore normal locomotive behaviors compared to controls. The NMJs in these larvae show a clear re-elaboration of motoneuron terminals with the formation of new synaptic branches and the addition of new synaptic boutons. Feeding newly eclosed flies RU486 results in robust rescue of climbing ability in TBPHΔ23 mutant flies.

Expression of TBPHScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4D42 significantly rescues the reduction in bouton number and terminal branches seen in muscles 6 and 7 of homozygous TBPHΔ23 third instar larvae. The lack of stable microtubules in TBPHΔ23 mutant distal boutons is also rescued.

Expression of TBPHScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4D42 rescues the defects in microtubule stability seen in TBPHΔ142 mutant distal boutons.

Expression of TBPHScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4elav.PU significantly rescues the reduction in bouton number and terminal branches seen in muscles 6 and 7 of homozygous TBPHΔ23 third instar larvae.
