Circadian entrained flies expressing Atx2KK100423 under the control of Scer\GAL4tim.UAS show decreased rhythmicity upon shift to constant darkness conditions.

Expression of Atx2KK100423 under the control of Scer\GAL4cry.PE does not significantly affect the rhythmicity of adult circadian locomotor activity under free-running (~constant darkness) conditions.

Expression of Atx2KK100423 (co-expressed with Dcr-2Scer\UAS.cDa) under the control of Scer\GAL4Mz520 results in a long-period locomotor rhythm in constant dark conditions. Loss of rhythmicity is also seen when Atx2KK100423 and Dcr-2Scer\UAS.cDa expressed under the control of Scer\GAL4P2.4.Pdf.

When Atx2KK100423 is expressed in all circadian neurons under the control of Scer\GAL4tim.PE (either with or without Dcr-2Scer\UAS.cDa) the period of circadian locomotor darkness is lengthened to ~27 hours. A similar increase is seen when Atx2KK100423 is expressed under the control of Scer\GAL4P2.4.Pdf. No phenotype is observed when Atx2KK100423 is expressed under the control of Scer\GAL4tim.PE in combination with Scer\GAL80Pdf.PS to block transcription in Pdf cells.

Circadian entrained flies co-expressing Atx2KK100423 and Nab2KK101974 under the control of Scer\GAL4tim.UAS show increased circadian period and decreased rhythmicity power upon shift to constant darkness conditions.

The poor rhythmicity of adult circadian locomotor activity under free-running (~constant darkness) conditions observed in flies expressing me31BNIG.4916R under the control of Scer\GAL4cry.PE is further worsened by co-expression of Atx2KK100423.

Knocking down Atx2 expression (using Atx2KK100423 under the control of Scer\GAL4tim.PE) in a tyfΔ11 mutant background results in a similar increase in period length as is seen in either mutant alone.

Expression of perScer\UAS.cYa suppresses the lengthening of the circadian locomotor darkness period seen when Atx2KK100423 is expressed under the control of Scer\GAL4tim.PE.
