Expression of cazKK107486 under the control of Scer\GAL4nSyb.PU does not cause developmental lethality or locomotor defects in a climbing assay. Co-expression of cazKK107486 and cazHMS00790 or cazKK107486 and cazHMS00156 under the control of Scer\GAL4nSyb.PU does not cause developmental lethality, but co-expression of cazKK107486 and cazHMS00790 under the control of Scer\GAL4nSyb.PU does result in modest climbing defects.

Knockdown of caz through the expression of cazKK107486 in the developing eye in the region posterior to the morphogenetic furrow, under the control of Scer\GAL4GMR.PS, induces morphologically aberrant rough eyes. SEM images show fusion of ommatidia and a lack of bristles.

caz knockdown eye discs (generated through expression of cazKK107486 under the control of Scer\GAL4GMR.PS) exhibit wild-type photoreceptor cells and neurons. There appears to be some increase in the occurrence of cone cell fusion. The orientation of ommatidia in these mutants can be irregular, and the size of ommatidia can vary. In addition, some ommatidia are apparently fused, suggesting that knockdown of caz disrupts differentiation of pupal ommatidial cell types, especially cone cells, and 1[o], 2[o], and 3[o] pigment cells.

Flies expressing cazKK107486 under the control of Scer\GAL4elav.PLu show no significant difference in life span compared to control flies. The flies show reduced ability in a climbing assay compared to controls.

The total length of synaptic branches and the number of boutons at the neuromuscular junction are reduced compared to wild type in larvae expressing cazKK107486 under the control of Scer\GAL4elav.PLu, while bouton size is normal.

Expression of cazKK107486 (co-expressed with Dcr-2Scer\UAS.cDa) under the control of Scer\GAL4Bx-MS1096 disrupts wing growth.

Ubiquitous expression of cazKK107486 (co-expressed with Dcr-2Scer\UAS.cDa) under the control of Scer\GAL4Act5C.PU reduces adult viability to approximately 30% at 25[o]C, but this is increased to approximately 60% when flies are raised at 19[o]C.

Expression of cazKK107486 (co-expressed with Dcr-2Scer\UAS.cDa) in neurons under the control of Scer\GAL4elav.PU severely reduces adult viability to approximately 25% at 25[o]C. Viability is increased to approximately 80% when flies are raised at 19[o]C.

Expression of cazKK107486 (co-expressed with Dcr-2Scer\UAS.cDa) in neurons under the control of Scer\GAL4D42 severely reduces adult viability to approximately 30% at 25[o]C. Viability is increased to approximately 75% when flies are raised at 19[o]C.

Expression of cazKK107486 (co-expressed with Dcr-2Scer\UAS.cDa) under the control of Scer\GAL4GMR.PF disrupts ommatidium formation in the larval eye disc.

Expression of cazKK107486 (co-expressed with Dcr-2Scer\UAS.cDa) under the control of Scer\GAL4VGlut-OK371 disrupts the arrangement of motor neurons in neuromeres A1-A5 of the third instar larval ventral nerve cord. Nuclear staining appears similar to wild type and no apoptosis is seen.

Down-regulation of caz through expression of cazKK107486 in the developing eye under the control of Scer\GAL4GMR.PF does not affect eye morphology. No locomotive defects are seen in flies expressing cazKK107486 in motorneurons under the control of Scer\GAL4VGlut-OK371.

Overexpression of BacA\p35Scer\UAS.cHa rescues the fusion of retinal cone cells induced by cazKK107486 expression.
