The expression of ChTKK109385 under the control of Scer\GAL4NP1131, Scer\GAL4c305a or Scer\GAL4Mef2.247 leads to third instar foraging larvae showing increased naive chemotactic response as well as decreased olfactory habituation, to ethyl acetate, amyl acetate or 3-Octanol; unlike controls, these larvae also are unable to dishabituate to ethyl acetate upon cold shock. Scer\GAL4NP1131- or Scer\GAL4Mef2.247-driven expression restricted to the third instar larval stage by Gal80[ts] induces the same responses to ethyl acetate.

The expression of ChTKK109385 under the control of Scer\GAL4Orco.2.642.Hsim\VP22 leads to third instar foraging larvae showing decreased naive chemotactic response to ethyl acetate.

The expression of ChTKK109385 under the control of Scer\GAL4GH298 leads to third instar foraging larvae showing decreased olfactory habituation to ethyl acetate; expression restricted to the third instar larval stage by Gal80[ts] induces increased naive chemotactic response as well as decreased olfactory habituation to ethyl acetate.

The expression of ChTKK109385 under the control of Scer\GAL4GH146 or Scer\GAL4NP2426 leads to third instar foraging larvae showing decreased olfactory habituation to ethyl acetate; expression restricted to the third instar larval stage by Gal80[ts] induces the same phenotype.

Scer\GAL4[ChAT.7.4]@ leads to third instar foraging larvae showing decreased naive chemotactic response and decreased olfactory habituation to ethyl acetate; expression restricted to the third instar larval stage by Gal80[ts] induces increased naive chemotactic response to ethyl acetate.

The expression of ChTKK109385 under the control of Scer\GAL4nSyb.PS induces a severe decrease in survival to pupation, with most individuals dying as embryos and the surviving larvae being severely less motile, as compared to controls; anyways, most pupae progress to adulthood. Expression under the control of Scer\GAL4nSyb.PS induces lower penetrances of the same phenotypes and also affects larval neuromuscular junctions, which show a significantly more branches, but not boutons, as compared to controls.

Individuals expressing ChTKK109385 under the control of Scer\GAL4Tab2-201Y develop normally throughout the larval and pupal stages but most fail to eclose from the pupal case; flies are alive till more than P14 stage but some are unable to push themselves out of the pupal case; adult escapies display severe flight and motor defects, and die within 1-2 days; adults also show abnormal abdominal melanization and wing expansion failure. Larvae show significantly decreased crawling ability (i.e. decreased peristaltic movements) and their neuromuscular junctions show significant increases in the numbers of boutons and branches, as compared to controls.

Whereas the Scer\GAL4repo-driven expression induces a complete eclosion failure, the Scer\GAL4c305a-driven expression does not induce eclosion defects, as compared to controls. The Scer\GAL4Mef2.247-driven expression does not induce any obvious eclosion defects or adult locomotion defects (i.e. climbing defects), as compared to controls. The Scer\GAL4ChAT.7.4-driven expression leads to decreased larval crawling ability (i.e. fewer peristaltic movements), but does not induce any obvious neuromuscular junction defects (i.e. in bouton or branch number) or eclosion defects, as compared to controls.

Scer\GAL4NP6024-driven expression leads to small but significant decreases in viability to pupation and in eclosing from the pupal case, as compared to controls; larvae show significantly decreased crawling ability (i.e. decreased peristaltic movements) and significant increases in the numbers of branches and boutons at the neuromuscular junctions, as compared to controls. Similarly, Scer\GAL4NP1131-driven expression induces a moderate decrease in viability to pupation and a small but significant decrease in eclosion from the pupal case, as compared to controls; larvae also show significantly decreased crawling ability (i.e. decreased peristaltic movements) and significant increases in the numbers of branches and boutons at the neuromuscular junctions, as compared to controls.

The eclosion defects induced when the expression of ChTKK109385 is under the control of Scer\GAL4Tab2-201Y, but not when under the control of Scer\GAL4repo, are suppressed by the co-expression of ChTUAS.cHa. The larval locomotion and neuromuscular junction defects induced by the Scer\GAL4Tab2-201Y-driven expression are also rescued by the co-expression of ChTUAS.cHa.

The larval locomotion and neuromuscular defects, the increased mortality to pupation and the eclosion defects induced by the expression of ChTKK109385 under the control of either Scer\GAL4NP1131 or Scer\GAL4NP6024 are partially rescued by the co-expression of ChTUAS.cHa.

The decreased larval locomotion induced by the expression of ChTKK109385 under the control of Scer\GAL4ChAT.7.4 is rescued by the co-expression of ChTUAS.cHa.

The larval neuromuscular junction defects induced by the expression of ChTKK109385 under the control of Scer\GAL4nSyb.PS are rescued by the co-expression of ChTUAS.cHa.
