UASt regulatory sequences drive expression of an inverted repeat.
Expression of Inx7KK112684 under the control of Scer\GAL4ey-OK107 in adults does not significantly affect 3 hour aversive olfactory memory or the residual anaesthesia-resistant memory (ARM) after cold-shock, compared to controls; expression under the control of Scer\GAL4VT000765 also does not affect ARM, compared to controls.
Expression of Inx7KK112684 under the control of Scer\GAL417d, the split-GAL4 pair of Scer\GAL4DBD.R41C07 and Hsap\RELAAD.R13F02, the split-GAL4 pair of Scer\GAL4DBD.R89B01 and Hsap\RELAAD.R13F02, or the split-GAL4 pair of Scer\GAL4DBD.R14C08 and Hsap\RELAAD.R12C11, does not affect visual short term memory following training with pain stimulus in adult flies, compared to controls.
Expression of Inx7KK112684 under the control of Scer\GAL4tj.PU (along with expression of UAS-Dcr2 to increase RNAi efficiency) does not cause sterility.
Expression of inx7KK112684 in both the anterior paired lateral (APL) and dorsal paired medial (DPM) neurons in the mushroom body (using the Scer\GAL4GH146 and Scer\GAL4c316 drivers simultaneously) results in impaired 3 hour (intermediate-term) memory after associative conditioning compared to controls, although initial learning is normal in these flies. The anesthesia-resistant memory (ARM) component of intermediate-term memory is normal in these flies.
Impairment of 3 hour memory after associative conditioning is still seen in flies in which expression of inx7KK112684 using the Scer\GAL4GH146 and Scer\GAL4c316 drivers simultaneously is limited to the adult stage by using the temperature sensitive Scer\GAL80ts.αTub84B allele to restrict expression before this stage.
The gross anatomy of the APL and DPM neurons in flies expressing inx7KK112684 using the Scer\GAL4GH146 and Scer\GAL4c316 drivers simultaneously is not different from that in control flies.
Flies expressing inx7KK112684 in the DPM neurons under the control of Scer\GAL45015 have normal 3 hour memory after associative conditioning.
Flies expressing inx7KK112684 under the control of Scer\GAL4GH146 have impaired 3 hour memory after associative conditioning compared to controls. If the expression is refined by adding the Scer\GAL80Cha.PK repressor (this reduces expression in the APL neurons to undetectable levels, leaving expression in only a subset of projection neurons), the flies have normal 3 hour memory.
