Expression of Zasp66KK112973 under the control of Scer\GAL4Mef2.PR results in 83% pupal lethality and surviving adults can fly. Mild defects in Z-disc structure are seen in the indirect flight muscles (IFMs) of adult flies.

Expression of Zasp66KK112973 under the control of Scer\GAL4Mef2.PR (in the presence of Dcr-2Scer\UAS.cDa to enhance RNAi) results in 98% pupal lethality and adults escapers are flightless. Mild myofibrillar abnormalities are seen in the IFMs of most adult escapers and distorted M-lines and Z-discs are occasionally seen. In more severe cases, sarcomere integrity is lost and M-lines and Z-discs are severely disrupted. At 48 hours after puparium formation the IFM myofibrils are thinner than normal and frayed and Z-discs are irregular.

Adults expressing Zasp66KK112973 under the control of Scer\GAL4Act88F.PB are able to fly and the indirect flight muscles show no severe defects ultrastructurally.

Adults co-expressing Zasp52KK101276 and Zasp66KK112973 under the control of Scer\GAL4Act88F.PB have severe defects in the indirect flight muscles. Myofibrils are often frayed and unstable, with severely distorted Z-discs and H-zones.
