Third instar larvae expressing ykiS111A.S168A.S250A.Scer\UAS.T:SV5\V5 under the control of Scer\GAL4RapGAP1-OK6 show normal number of satellite boutons on neuromuscular junctions relative to controls.
Expression of ykiS111A.S168A.S250A.Scer\UAS.T:SV5\V5 in adult midgut intestinal stem cells (under the control of Scer\GAL4esg-NP5130 and limited to the adult stage using Scer\GAL80ts.αTub84B) results in massive cell proliferation resulting in a bloating phenotype. This progressive phenotype is seen from 5 days after ykiS111A.S168A.S250A.Scer\UAS.T:SV5\V5 induction onwards. The ovaries of these flies degenerate progressively, and in contrast to the continuous mass seen in wild type flies the fat bodies of flies expressing ykiS111A.S168A.S250A.Scer\UAS.T:SV5\V5 appear as discontinuous patches that later degenerate completely. Progressive climbing defects and downturned wings are also seen. The mitochondria of muscles expressing ykiS111A.S168A.S250A.Scer\UAS.T:SV5\V5 are swollen and often associated with empty space. Mitochondrial degeneration is seen. The volume of extractable hemolymph from flies expressing ykiS111A.S168A.S250A.Scer\UAS.T:SV5\V5 in adult midgut intestinal stem cells (under the control of Scer\GAL4esg-NP5130 and limited to the adult stage using Scer\GAL80ts.αTub84B) is greatly increased compared to controls. The concentration of trehalose in the hemolymph is significantly increased. Whole-body trehalose levels are increased. Whole-body triglycerides and glycogen are reduced compared to controls. Food intake and excretion are unaffected. Increased glucose incorporation is seen in the midgut of flies expressing ykiS111A.S168A.S250A.Scer\UAS.T:SV5\V5 in adult midgut intestinal stem cells (under the control of Scer\GAL4esg-NP5130 and limited to the adult stage using Scer\GAL80ts.αTub84B) compared with controls. In other parts of these flies, glucose incorporation is reduced.
Scer\GAL4ey.PH-mediated expression results in tumourous eyes.
The heart walls of flies expressing ykiS111A.S168A.S250A.Scer\UAS.T:SV5\V5 under the control of Scer\GAL4tin.CΔ4 are thicker than controls but cardiomyocyte number is the same as in controls.
When ykiS111A.S168A.S250A.Scer\UAS.T:SV5\V5 is expressed under the control of Scer\GAL4esg-NP5130 and Scer\GAL80ts.αTub84B, midgut stem cells over-proliferate.

Expression of ykiS111A.S168A.S250A.Scer\UAS.T:SV5\V5 under the control of Scer\GAL4sd-SG29.1 results in early larval lethality. Flies carrying ykiS111A.S168A.S250A.Scer\UAS.T:SV5\V5 in the presence of Scer\GAL4GMR.PF and Scer\GAL80αTub84B.PL show obvious eye overgrowth.
