Scer\GAL4esg-NP5130-mediated expression of Src64BScer\UAS.cCa (restricted to adult stages by using P{tubP-GAL80ts}) results in substantial intestinal stem cell hyperproliferation and hyperplasia of the adult intestine.

Expression of CskdsRNA.Scer\UAS (co-expressed with Dcr-2Scer\UAS.cDa) under the control of Scer\GAL4ptc-559.1 results in mild but consistent cell migration from the ptc domain of the wing disc compared to controls.

Expression of CskIR.Scer\UAS using Scer\GAL4unspecified causes a weak mispatterning phenotype in the adult eye.

Expression of CskIR.Scer\UAS under the control of Scer\GAL4ptc-559.1 in the wing disc resulting in the delamination, movement and apoptosis of some of the mutant cells along the boundary between the mutant and wild-type cells.

CskIR.Scer\UAS; Scer\GAL4GMR.PF adults have enlarged, rough eyes. Analysis of the developing retinas of these animals at 42 hours after puparium formation shows an excess and mis-patterning of secondary and tertiary pigment cells. Even in areas of the retina with normal numbers of these cells, their arrangement is disrupted. Earlier still, at 29 hours after puparium formation, there is a striking reduction in apoptosis in the developing retina. In particular, excess inter-ommatidial precursor cells fail to die. Initially these precursor cells mostly take up a normal arrangement in the inter-ommatidial lattice, but they then fail to hold these positions - often sliding over each other to form piles rather than remaining as a monolayer.

CskIR.Scer\UAS; Scer\GAL4Act5C.PI animals die as young pupae with a body size approximately 50% larger than wild-type controls.

CskIR.Scer\UAS; Scer\GAL4C-765 adults have significantly larger wings than controls and have extra vein material in veins L2 and L5 and the posterior crossvein.

CskIR.Scer\UAS; Scer\GAL4Act5C.PI somatic clones in the eye disc are smaller than their wild-type twin spots and rarely survive in the pupal retina. Cell death in these clones in concentrated at their boundaries.

The wings of CskIR.Scer\UAS; Scer\GAL4sd-SG29.1 adults are severely reduced and malformed. In contrast, the wings of CskIR.Scer\UAS; Scer\GAL4ptc-559.1 adults are normal in size, but have ectopic vein material associated with wing vein L4 and a missing or defective anterior crossvein. The wing discs of these animals contain increased numbers of apoptotic cells at the posterior edge of the ptc expression domain. In addition, cells in a region extending 3-4 cell diameters anterior to the anterior-posterior boundary lose components of their zonula adherens and then lose their apical profiles. These cells often delaminate completely from the wing disc epithelium and some end up on the posterior side of the compartment boundary, despite their origins in the anterior compartment. No increase in cell death or loss of apical profiles is seen in CskIR.Scer\UAS; Scer\GAL4bi-omb-Gal4 wing discs.

Co-expression of CskdsRNA.Scer\UAS and Sin3AKK100700 (with Dcr-2Scer\UAS.cDa) results in a much stronger wing disc cell migration phenotype than is seen when either transgene is expressed alone. Upon co-expression of bskK53R.Scer\UAS, massive overgrowth is seen, but the invasion phenotype is suppressed.

The mispatterning seen in CskIR.Scer\UAS, Scer\GAL4unspecified eyes is: (i) restored when eyes are also made homozygous mutant for Dcr-2unspecified, or (ii) strongly enhanced when eyes are also made homozygous mutant for HPS4unspecified (using the "eyFLP/FRT" system).

ASPP8/ASPP8 dramatically enhances the delamination, movement and apoptosis phenotype seen in Csk mutant cells which are at the boundary between mutant and wild-type cells in wing discs expressing CskIR.Scer\UAS under the control of Scer\GAL4ptc-559.1.

CskIR.Scer\UAS; Scer\GAL4GMR.PF partially suppresses the reduction in eye size due to rprGMR.PW.

The extra secondary and tertiary pigment cell phenotype seen in the developing retinas of CskIR.Scer\UAS; Scer\GAL4GMR.PF animals at 42 hours after puparium formation is suppressed by shgScer\UAS.cSa.

Rho172O/+ or pucScer\UAS.cMa or shgR69/+ suppress the loss of apical profile, delamination and subsequent migration and cell death increase seen in cells at the posterior edge of theptc expression domain in CskIR.Scer\UAS Scer\GAL4ptc-559.1 third instar larvae. p120ctn308/+ partially suppresses these phenotypes. In contrast, pucE69/+ strongly enhances these phenotypes.
