Expression of HipkUAS.Tag:HA under the control of Scer\GAL4dpp.blk1 leads to overproliferation and cell spreading of Hipk[UAS. Tag:HA ] expressing cells in third instar larval wing discs compared to controls.

The expression of HipkUAS.Tag:HA under the control of Scer\GAL4dpp.blk1 at 25[o]C induces dramatic overgrowth of eye, wing and leg imaginal discs, characterized by expansion of the Dpp expression domain and tissue folds and protrusions; at 29[o]C, this expression induces overgrown wing discs that show apoptosis (cleaved Caspase 3 staining) within the Dpp expression domain. Unlike controls, a) clusters of pigmented retinal cells are occasionally found in the adult thorax, b) eye disc cells extrude from the disc and continue to proliferate after leaving the disc, c) cells originating from the wing disc's Dpp-positive domain are found towards both anterior and posterior parts of disc, d) wing disc cells appear to intercalate into the basement membrane and extrude through holes in the basement membrane.

Expression of HipkUAS.Tag:HA under the control of Scer\GAL4dpp.blk1 and Scer\GAL4Hml.Δ induces melanotic tumors in larvae and adults, respectively.

The expression of HipkUAS.Tag:HA under the control of Scer\GAL4ey.PH occasionally leads to a cluster of pigmented retinal cells (from the ey domain in the eye) being observed in the adult thorax, despite of no obvious eye defects. Unlike control eye discs, which show a tight boundary of photoreceptor cells at the posterior margin, this expression alters cell morphology, with jagged cell extensions that protrude towards the posterior margin of the disc.

Clonal expression of HipkUAS.Tag:HA under the control of Scer\GAL4Act5C.PP in the wing disc induces apoptosis (cleaved Caspase 3 staining) in a cell autonomous manner.

Expression of hipkScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4bi.PU results in dosage-dependent overgrowth in third instar larval wing discs.

Expression of hipkScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4ey.PH at 29[o]C results in rough eyes, of which 33% also display cuticle-like structures. 39% of these flies have overgrown eyes and overgrowths of eye discs are also seen in third instar larvae.

Expression of one copy of hipkScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4bi.PU results in the formation of an additional wing margin and outgrowths from the ventral side of the wing. Expression of two copies of hipkScer\UAS.T:Ivir\HA1 results in the formation of additional wing tissue outgrowths.

Ectopic expression of hipkScer\UAS.T:Ivir\HA1 in the wing using Scer\GAL469B results in ectopic vein formation.

Co-expression of hipkScer\UAS.T:Ivir\HA1 and ykiScer\UAS.T:Avic\GFP-EGFP,T:Ivir\HA1 under the control of Scer\GAL4hh-Gal4 enhances the wing disc overgrowth seen when either construct is expressed alone.

Expression of hipkScer\UAS.T:Ivir\HA1 suppresses the overgrowth seen when ykiS168A.Scer\UAS.T:Avic\GFP-EGFP,T:Ivir\HA1 is expressed under the control of Scer\GAL4ptc-559.1. The phenotype is rescued in all tissues examined, including the wing disc and the eye. Fewer ykiS168A.Scer\UAS.T:Avic\GFP-EGFP,T:Ivir\HA1 expressing cells are seen, indicating that cell proliferation has been reduced.

The eye overproliferation defect caused by expression of DlScer\UAS.cDa under the control of Scer\GAL4ey.PH is enhanced by co-expression of hipkScer\UAS.T:Ivir\HA1.

The eye loss phenotype caused by expression of NECN.Scer\UAS.cGa under the control of Scer\GAL4ey.PH is partially suppressed by co-expression of hipkScer\UAS.T:Ivir\HA1.

Co-expression of hipkScer\UAS.T:Ivir\HA1 partially suppresses the eye loss phenotype caused by expression of groScer\UAS.cAa under the control of Scer\GAL4ey.PH.

Co-expression of hipkScer\UAS.T:Ivir\HA1 does not suppress the eye loss phenotype caused by expression of groAA.cLa.Scer\UAS under the control of Scer\GAL4ey.PH.

Co-expression of hipkScer\UAS.T:Ivir\HA1 partially suppresses the complete eye loss caused by expression of HScer\UAS.cGa under the control of Scer\GAL4ey.PH; a small eye is seen.

Co-expression of hipkScer\UAS.T:Ivir\HA1 partially rescues the small eye phenotype caused by expression of fngScer\UAS.cKa under the control of Scer\GAL4ey.PH.

Expression of hipkScer\UAS.T:Ivir\HA1 under the control of Scer\GAL469B in a wg1/wgl-17 mutant background results in proper wing development and rescues the wing-to-notum transformation.

Expression of hipkScer\UAS.T:Ivir\HA1 in animals expressing fz2Scer\UAS.N under the control of Scer\GAL469B rescues the severely notched wing phenotype.

Individuals co-expressing BacA\p35UAS.cHa and HipkUAS.Tag:HA under the control of Scer\GAL4dpp.blk1 arrest at the third larval stage for an extended period of time (beyond 10 days) and eventually die as larvae. BacA\p35UAS.cHa co-expression enhances the wing disc tumorigenesis of the expression of HipkUAS.Tag:HA expression (the Dpp expression domain cells expand substantially and occupy almost the entire disc) and does not suppress the melanotic masses.
