dlg1A40.2 larval wing discs exhibit tumors, as well as evident apoptosis (Dcp1 staining).

dlg1A40.2 hemizygotes show wing disc overgrowth, associated with a significant increase in cell number, as compared to controls.

The wing discs of dlg1A40.2 hemizygous third instar larvae show epithelial cell polarity defects and hyperplasia, as compared to controls.

dlg12/dlg1A40.2 transheterozygosity leads to the formation of large tumors in the third instar larval wing disc.

dlg1A40.2 mutant larvae display wing disc tumors and a severe increase in the density of circulating hemocytes, as compared to controls.

dlg1A40.2 homozygous larvae have an enlarged brain, and tumorous imaginal discs.

There is a significant increase in the length of each pre-synaptic density at larval neuromuscular junctions in dlg1A40.2 homozygotes compared to wild-type.

Electrophysiological readings from the neuromuscular junctions of dlg1A40.2 homozygotes show significantly decreased evoked excitatory junction potential amplitude compared to controls.

imd1, RelE20, DefSK3 or DefSK4 enhance the tumor phenotype and suppress the associated apoptosis (Dcp1 staining) observed in dlg1A40.2 wing discs; the same happens upon the expression of DefKK111656 under the control of either Scer\GAL4αTub84B.PL, Scer\GAL4apolpp.PB or Scer\GAL4btl.PS, upon the expression of imdKK109863 under the control of Scer\GAL4btl.PS or Scer\GAL4apolpp.PB, or upon the expression of Myd88GD9716 under the control of Scer\GAL4apolpp.PB. DefSK3 and DefSK4 do not affect the cell division rate in dlg1A40.2 wing discs.

Scer\GAL4apolpp.PB-driven expression of DefUAS.cTa in DefSK3, dlg1A40.2 double mutants, imdKK109863, dlg1A40.2 double mutants, or Myd88GD9716, dlg1A40.2 double mutants partially re-establishes the original dlg1A40.2 phenotypes.

Injecting synthetic Defensin protein into dlg1A40.2 larvae enhances the apoptosis observed in their wing discs. Injecting synthetic Defensin protein into DefSK3, dlg1A40.2 and egr3, dlg1A40.2 larvae re-establishes the increased apoptosis observed in dlg1A40.2 larvae.

The tumor phenotype of dlg1A40.2/dlg12 transheterozygous third instar larval wing discs is partially suppressed by the expression of Socs36EScer\UAS.P\T.cCa under the control of Scer\GAL4en-e16E, as it leads to a significantly decreased cell number in the discs' posterior compartment, and is enhanced by the expression of either ph-pGD4480 or Su(z)2KK103634 under the control of Scer\GAL4en-e16E, as they lead to a significantly increased cell number in the discs' posterior compartment.

The apoptosis observed in dlg1A40.2 mutant wing disc tumors is virtually abrogated by the expression of egrKK103432, alone or in combination with Ras85DV12.UAS, by the expression of PvrKK101575 alone or by the expression of spzKK112908 alone, is significantly decreased by the expression of grimUAS.cUa, is significantly increased by the expression of egrUAS.Venus, but does not seem to be affected by the expression of Ras85DV12.UAS alone (all expression conditions are under the control of the Scer\GAL4Hml.Δ driver).

The apoptosis observed in dlg1A40.2 mutant wing disc tumors is also virtually abrogated by the expression of spzKK112908 under the control of Scer\GAL4He.PZ, by the expression of Myd88VDRC.cUa under the control of either Scer\GAL4ppl.PP, Scer\GAL4Cg.PA or Scer\GAL4Lsp2.PH, or by the co-expression of TlUASp.Venus and egrKK103432 under the control of Scer\GAL4Cg.PA, while being significantly increased by the expression of TlUASp.Venus under the control of either Scer\GAL4ppl.PP or Scer\GAL4Cg.PA.
