Flies expressing Pld[Scer\UAS.cRa] under the control of Scer\GAL4[GMR.PF] show vesiculation and degeneration of the rhabdomeres. In addition, the cell body of the photoreceptors shows accumulation of expanded tubulovesicular membraneous structures, which appear as concentric stacks of membranes in the most severe cases. Retinal extracts from flies expressing Pld[Scer\UAS.cRa] under the control of Scer\GAL4[GMR.PF] show a significant elevation of a single species of phosphatidic acid (predicted fatty acyl composition 16:0/18:2) compared to controls. The levels of phosphatidylcholine and phosphatidylinositol in the retinal extracts are normal.

Co-expression of CG11426[Scer\UAS.T:Avic\GFP] partially rescues the rhabdomere degeneration caused by expression of Pld[Scer\UAS.cRa] under the control of Scer\GAL4[GMR.PF]. Co-expression of garz[EP2028] strongly enhances the retina phenotype caused by expression of Pld[Scer\UAS.cRa] under the control of Scer\GAL4[GMR.PF]; only one or two poorly developed rhabdomeres are seen. Co-expression of Gap69C[Scer\UAS.cRa] partially the rhabdomere defects caused by expression of Pld[Scer\UAS.cRa] under the control of Scer\GAL4[GMR.PF]. Co-expression of siz[P2.Scer\UAS] does not enhance the photoreceptor cell phenotypes caused by expression of Pld[Scer\UAS.cRa] under the control of Scer\GAL4[GMR.PF]. Arf51F[4] does not suppress the photoreceptor cell phenotypes caused by expression of Pld[Scer\UAS.cRa] under the control of Scer\GAL4[GMR.PF]. Animals co-expressing both Pld[Scer\UAS.cRa] and α-Adaptin[dsRNA.Scer\UAS.cUa] under the control of Scer\GAL4[GMR.PF] have virtually no detectable rhabdomeres, and the cell bodies of the photoreceptors continue to show accumulation of membrane transport intermediates. A shi[1] background at 23[o]C does not suppress the rhabdomere defects caused by expression of Pld[Scer\UAS.cRa] under the control of Scer\GAL4[GMR.PF]. There is a dramatic increase in rhabdomere size in flies expressing Pld[Scer\UAS.cRa] under the control of Scer\GAL4[GMR.PF] in a sktl[Δ1-1]/sktl[Δ20] background compared to flies expressing Pld[Scer\UAS.cRa] under the control of Scer\GAL4[GMR.PF] in a wild-type background.
