Description

P{EP} element insertion approximately 3kb upstream of the nej open reading frame, in the 5' UTR. The orientation of the P{EP} element is such that the 3' end of the P{EP} element is proximal to the nej open reading frame, allowing overexpression of nej under the control of the Scer\UAS regulatory sequences present in the P{EP} element.

Third instar larvae expressing nejEP1149 under the control of Scer\GAL4Pxn.PS have a decreased number of sessile hemocytes. The number of hemocytes in circulation is also decreased. The dorsal sessile hemocyte department is disrupted. Sessile hemocytes accumulate along the dorsal vessel but do not spread through the cuticle. The lymph gland is enlarged. Most hemocytes appear wild type in shape, although a few larger cells can also be detected.

Expression of nejEP1149 under the control of Scer\GAL4GMR.PF results in a small eye phenotype.

Expression of CG15321EP1149 under the control of Scer\GAL4ey.PH results in eyes that are strongly reduced in size.

Expression of nejEP1149 under the control of Scer\GAL4sca-537.4 results in potential socket-to-shaft and shaft-to-socket transformations, severe loss of scutellar and dorsocentral macrochaetae, short and thickened shaft morphology and sockets with shaft-like protrusions.

nejEP1149/nejTA57 or nejEP1149/nejS342 transheterozygotes are semi-viable as third instar larvae. These larvae are developmentally delayed by approximately 1 day and are sluggish. Homozygous larvae show a significant decrease in presynaptic transmitter release (quantal content) without any change in quantal size at the neuromuscular junction. No change in the average muscle resting potential or in the average muscle input resistance is seen. Synaptic ultrastructure at neuromuscular boutons appears normal. There is an increase in the number of presynaptic T bars per active zone compared to wild type. Overexpression of nejEP1149 under the control of Scer\GAL4elav.PLu results in early larval lethality. Overexpression of nejEP1149 under the control of Scer\GAL4elav.PLu (in nejEP1149 heterozygotes) results in a decrease in presynaptic transmitter release at the neuromuscular junction, without any change quantal size. There is a significant (although moderate) increase in the number of boutons at the neuromuscular junction of muscles 4, 6 and 7 compared to wild type in nejEP1149/nejS342, nejEP1149/nejQ7 or nejEP1149/nejTA57 larvae. No alteration in axon guidance in the embryonic central nervous system is seen.

nejEP1149 fails to complement nejTA57 and nejS342 for synaptic transmission defects. The synaptic defects seen at the neuromuscular junction in hemizygous nejEP1149 larvae are rescued when nejEP1149 is overexpressed using Scer\GAL4Mhc.PW.
