Expression of Inx2JF02446 under the control of Scer\GAL4ey-OK107 in adults does not significantly affect 3 hour aversive olfactory memory, compared to controls.

Expression of Inx2JF02446 under the control of Scer\GAL4repo.PL (in combination with tub-Gal80[ts] transgene to restrict expression to the adult stage) leads to a significantly reduced life span under sugar only diet when compared to controls.

Expression of Inx2JF02446 under the control of Scer\GAL4tj.PU (along with expression of UAS-Dcr2 to increase RNAi efficiency) results in disruption of spermatogenesis, sterility and small, rudimentary testes. Germline stem cell number in testes is significantly reduced, early somatic cell number is increased, and late somatic cell number is decreased. Somatic cyst stem cell proliferation is disrupted. The number of germline cysts that reach meiotic stages is reduced. Encapsulation of testis germline cysts occurs normally, and cadherin-mediated cell-cell adhesion is unaffected in testes expressing Inx2JF02446 under the control of Scer\GAL4tj.PU.

Neuronal expression of Inx2JF02446 under the control of Scer\GAL4elav-C155 does not affect fly viability or visual signal transmission.

Expression of Inx2JF02446 using Scer\GAL4Mz709 does not cause obvious morphological change to satellite glia. These flies show normal arrays of ommatidia and cartridges in the retina and lamina, respectively. In the lamina, fine structures such as capitate projections and T-bar synapses appear normal in photoreceptor terminals. However, photoreceptor terminals exhibit a reduced number of synaptic vesicles. The density of these synaptic vesicles is approximately 50% of wild-type.

Knockdown of Inx2 through expression of Inx2JF02446 using Scer\GAL4Mz709 results in the absence of ON and OFF transients in ERG recordings of the laminar LMC neurons at the onset and end of light stimulation.

Expression of Inx2JF02446 under the control of Scer\GAL4ptc-559.1 has no effect on the wing margin.

Expression of Inx2JF02446 and Inx3HM05245, both under the control of Scer\GAL4tj.PU, phenocopies expression of Inx2JF02446 alone under the control of Scer\GAL4tj.PU.

Co-expression of GmdNIG.8890R and Inx2JF02446 under the control of Scer\GAL4ptc-559.1 results in loss of wing margin.

Comments

Expression of Inx2Scer\UAS.T:Avic\GFP-EGFP in glia, under the control of Scer\GAL4repo, partially rescues flies expressing Inx2JF02446 under the control of Scer\GAL4repo. These rescued flies show normal visual transmission, excluding a role of neuronal Inx2 in visual transmission.
