Flies expressing ShawRA.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4tim.PE (but not Scer\GAL4qsm-NP6258 or Scer\GAL4qsm-NP6258) exhibit ultradian locomotor rhythms in constant dim white light, in contrast to control flies which largely show arrhythmic behavior in these conditions. These flies do not exhibit any difference in locomotor rhythms when kept in constant darkness, as compared to controls.

The l-LNv neurons in flies expressing ShawRA.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4Pdf.PU exhibit a hyperpolarized resting membrane potential, and a reduced spontaneous firing rate during the daytime, but not during the night, as compared to wild type; these neurons also fail to respond to blue light stimulus, in contrast to wild type.

Expression of ShawRA.Scer\UAS.T:Zzzz\FLAG in clock cells under the control of Scer\GAL4tim.PE causes flies to have more locomotor activity compared to controls. Activity is significantly increased at night. Adding Scer\GAL80Pdf.PS to remove overexpression in the LNv has no effect on the hyperactive phenotype. Restriction of expression to the Dorsal neurons (through the addition of Scer\GAL80cry.PS to Scer\GAL4tim.PE/ShawRA.Scer\UAS.T:Zzzz\FLAG has no effect on the increased night activity seen in Scer\GAL4tim.PE/ShawRA.Scer\UAS.T:Zzzz\FLAG mutants.

Expression of ShawRA.Scer\UAS.T:Zzzz\FLAG in clock cells under the control of Scer\GAL4cry.PE causes flies to have more locomotor activity compared to controls. Activity is significantly increased at night. The addition of Scer\GAL80Pdf.PS removes the locomotor activity increase found in Scer\GAL4cry.PE/ShawRA.Scer\UAS.T:Zzzz\FLAG overexpression mutants.

Expression of ShawRA.Scer\UAS.T:Zzzz\FLAG in clock cells under the control of Scer\GAL4P2.4.Pdf has no effect on locomotor activity compared to controls.

Expression of ShawRA.Scer\UAS.T:Zzzz\FLAG using Scer\GAL4tim.PE results in arrhythmic behavior immediately after the transition to DD. These flies also exhibit a reduced lifespan, with individuals never surviving until the end of an experiment (roughly surviving for 20 days or so).

Flies that express Shaw less extensively in the clock network (Scer\GAL4cry.PE/ShawRA.Scer\UAS.T:Zzzz\FLAG or Scer\GAL4P2.4.Pdf/ShawRA.Scer\UAS.T:Zzzz\FLAG) maintain rhythmicity in DD albeit with reduced amplitude compared to controls.

Expression of ShawRA.Scer\UAS.T:Zzzz\FLAG in the LNvs only (under the control of both Scer\GAL4tim.PE and Scer\GAL80Pdf.PS) appears to result in a small increase of rhythmically behaving flies compared to expressing ShawRA.Scer\UAS.T:Zzzz\FLAG in all clock neurons under the control of Scer\GAL4tim.PE.

Expression of ShawRA.Scer\UAS.T:Zzzz\FLAG in DN[[1]], DN[[2]] and DN[[3]] through Scer\GAL4tim.PE and Scer\GAL80cry.PS results in more than 30% of the progeny being arrhythmic in DD, with the rhythmic flies exhibiting rather weak rhythmicity.

Overexpression of ShawRA.Scer\UAS.T:Zzzz\FLAG using Scer\GAL4P2.4.Pdf results in a small decrease in sleep during both day and night, but an increase in sleep latency only during lights-off.

Some adults expressing ShawRA.Scer\UAS.T:Zzzz\FLAG with Scer\GAL4Ccap.PP show a phenotype with unfurled wings, softened or abnormally tanned cuticle, and show an ether-induced shaking phenotype. Some small pupae fail to eclose.

Firing properties and whole cell parameters of MNISN-Is motor neurons are not substantially different from controls in animals expressing ShawRA.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4C380.
