Mutant flies show greatly reduced survival compared to wild type after septic injury with Erwinia carotovora carotovora 15.
Iap2[7c]/Df(2R)Exel7138 or Iap2[7c] homozygous third instar larvae subjected to sublethal doses of x-ray irradiation (1.5 Gy) display a marked increase in wing imaginal disc apoptosis compared to wild-type larvae.
86% of Iap2[7c]/Df(2R)Exel7138 animals survive embryogenesis and develop normally, however animals are acutely sensitive to infections. Iap2[7c]/Df(2R)Exel7138 animals are highly susceptible to infection by gram-negative bacteria, but not to gram-positive bacteria and fungi. Iap2[7c]/Df(2R)Exel7138 flies die within 25 hours of septic injury with gram-negative bacteria, and almost identical susceptibility is observed for Iap2[7c], or Iap2[7c]/Iap2[7a] transheterozygous mutant animals.

Expression of Iap2[RINGmut.Scer\UAS] under the control of Scer\GAL4[da.G32] fails to rescue the reduced survival after septic injury with Erwinia carotovora carotovora which is seen in Iap2[7c] flies. Expression of Iap2[Scer\UAS.cLa] under the control of Scer\GAL4[da.G32] rescues the reduced survival after septic injury with Erwinia carotovora carotovora 15 which is seen in Iap2[7c] flies. Expression of either Iap2[Scer\UAS.attB], Iap2[BIR2mut.Scer\UAS] or Iap2[BIR3mut.Scer\UAS] under the control of Scer\GAL4[c564] rescues the reduced survival after septic injury with Erwinia carotovora carotovora 15 which is seen in Iap2[7c] flies. Expression of Iap2[BIR23mut.Scer\UAS] under the control of Scer\GAL4[da.G32] fails to rescue the reduced survival after septic injury with Erwinia carotovora carotovora 15 which is seen in Iap2[7c] flies.
Enhanced sensitivity of Iap2[7c] homozygotes to x-ray irradiation is rescued by Iap2[+t3.3].
Leaky, low-level expression of Iap2[Scer\UAS.cLa] in a Iap2[7c] mutant background is sufficient to completely rescue the immunodeficient phenotype observed in Iap2[7c] mutant animals in response to infection by gram-negative bacteria. Iap2[7c] mutant animals expressing Iap2[Scer\UAS.cLa], under the control of Scer\GAL4[Act5C] or Scer\GAL4[da.G32], are fully resistant to infection by gram-negative bacterium and survive microbial exposure at a similar rate to wild-type.
