nahar mutants exhibit defects in morning anticipatory behaviour and light-induced activity.

Mutants show a strong decrease in the rhythmic amplitude of circadian locomotor activity upon release into constant darkness. The activity distribution is also altered compared to wild type under constant darkness, with the mutant flies having a single broad activity peak across the subjective day (wild-type controls have a prominent peak that approximates to the phase of the evening peak seen in light-dark conditions and a less peak that approximates the morning peak seen in light-dark conditions). The mutant flies also show an increase in the period length of circadian locomotor activity under constant darkness conditions.

DN1[[P] rescue of nahar mutants through expression of naScer\UAS.cLa under the control of Scer\GAL4Clk.4.1M restores only the acute responsiveness to lights-on (AMI), but not the sustained masking index.

Morning anticipatory behaviour is restored in nahar mutants where naScer\UAS.cLa is expressed under the control of Scer\GAL4Clk.4.1M. However, evening behaviour under LD conditions is not rescued.

Expression of naScer\UAS.cLa under the control of either Scer\GAL4elav-C155, Scer\GAL4Clk.8.0 or Scer\GAL4tim.PE results in strong rescue of the circadian locomotor activity defects seen in nahar flies under 12 hour light/12 hour dark conditions, with rescue of evening anticipation, timing of morning anticipation and response to lights on.

Expression of naScer\UAS.cLa under the control of Scer\GAL4cry.PZ results in robust rescue of the circadian locomotor activity defects seen in nahar flies under 12 hour light/12 hour dark conditions.

Expression of naScer\UAS.cLa under the control of either Scer\GAL4P2.4.Pdf or Scer\GAL41118 fails to rescue the circadian locomotor activity defects seen in nahar flies under 12 hour light/12 hour dark conditions.

The rescue of the circadian locomotor activity defects seen in nahar flies under 12 hour light/12 hour dark conditions by expression of naScer\UAS.cLa under the control of Scer\GAL4elav-C155 is almost completely blocked if the flies are also expressing Scer\GAL80cry.PS.

The rescue of the circadian locomotor activity defects seen in nahar flies under 12 hour light/12 hour dark conditions by expression of naScer\UAS.cLa under the control of either Scer\GAL4elav-C155, Scer\GAL4Clk.8.0 or Scer\GAL4tim.PE is not strongly affected in the flies are also expressing Scer\GAL80Pdf.PS.

Expression of naScer\UAS.cLa under the control of either Scer\GAL4cry.PE or Scer\GAL4Mai179 partially rescues the circadian locomotor activity defects seen in nahar flies under 12 hour light/12 hour dark conditions; evening anticipation is rescued, but the lights-on activity response is not rescued. Morning anticipation is not affected if expression is driven by Scer\GAL4Mai179, but if expression is driven by Scer\GAL4cry.PE the degree of morning anticipation is suppressed compared to nahar mutant controls. Evening anticipation is still rescued in nahar expressing naScer\UAS.cLa under the control of Scer\GAL4cry.PE that are also expressing Scer\GAL80Pdf.PS.

Expression of naScer\UAS.cLa under the control of either Scer\GAL4elav-C155 or Scer\GAL4Clk.8.0 significantly rescues the circadian locomotor activity defects seen in nahar flies under constant darkness conditions, with a rescue of both morning and evening peaks being seen.

Expression of naScer\UAS.cLa under the control of Scer\GAL4cry.PE partially rescues the circadian locomotor activity defects seen in nahar flies under constant darkness conditions; rescue of a single broad evening peak is seen, but there is either no morning anticipatory behaviour or it is very delayed and there are no reductions in midday activity compared to nahar mutant controls.

Expression of naScer\UAS.cLa under the control of Scer\GAL4Mai179 partially rescues the circadian locomotor activity defects seen in nahar flies under constant darkness conditions; there is a strong rescue of the evening activity peak and a reduction in midday activity, but there is no change in morning anticipatory behaviour compared to nahar mutant controls.

Expression of naScer\UAS.cLa under the control of Scer\GAL4tim.PE significantly rescues the circadian locomotor activity defects seen in nahar flies under constant darkness conditions.

Expression of naScer\UAS.cLa under the control of Scer\GAL4cry.PZ weakly rescues the circadian locomotor activity defects seen in nahar flies under constant darkness conditions; a higher proportion of flies with rhythmic behaviour is seen and the rescued flies show clear persistant morning and evening peaks.

Expression of naScer\UAS.cLa under the control of Scer\GAL4P2.4.Pdf fails to rescue the circadian locomotor activity defects seen in nahar flies under constant darkness conditions.

The rescue of the circadian locomotor activity defects seen in nahar flies under constant darkness conditions by expression of naScer\UAS.cLa under the control of either Scer\GAL4elav-C155, Scer\GAL4Clk.8.0, Scer\GAL4cry.PE or Scer\GAL4tim.PE is reduced if the flies are also expressing Scer\GAL80Pdf.PS.

Expression of naE.Scer\UAS under the control of Scer\GAL4tim.PE fails to rescue the circadian locomotor activity defects seen in nahar flies under 12 hour light/12 hour dark conditions.

Expression of naE.Scer\UAS under the control of Scer\GAL4Clk.8.0 partially rescues the circadian locomotor activity defects seen in nahar flies under 12 hour light/12 hour dark conditions; there is no rescue of the timing of morning anticipatory behaviour, but there is partial rescue of evening anticipation.

Expression of naE.Scer\UAS under the control of Scer\GAL4Clk.8.0 does not generally rescue the the circadian locomotor activity defects seen in nahar flies under constant darkness conditions.
