Somatic clones expressing wupAJF02172 under the control of Scer\GAL4Act.PU in wandering third instar larval wing discs present a significant and progressive decrease in the relative clonal area, as it is smaller at 72h than at 48h after clonal induction, present significant apoptosis, and present recruitment of hemocytes to clonal cells, as compared to control clones.

The expression of wupAJF02172 under the control of Scer\GAL4en.PU leads to increased apoptosis in the posterior compartment in wandering third instar larval wing discs, as compared to controls.

Walking and jumping behaviors are unaffected in flies expressing wupAJF02172 under the control of Scer\GAL4Hk-UH3 (limited to the late stages of development using Scer\GAL80ts.αTub84B). Abnormalities are seen in the indirect flight muscle fascicles compared to the wild type and sarcomere organisation is disrupted. This becomes more severe when wupAJF02172 is co-expressed with Dcr-2.

The co-expression of either fweKK109356, dppScer\UAS.cUa.T:Avic\GFP, SPARCScer\UAS.cPa or bskDN.Scer\UAS suppresses the decreased relative clonal area of somatic clones expressing wupAJF02172 under the control of Scer\GAL4Act.PU in wandering third instar larval wing discs; the co-expression of fweKK109356 or SPARCScer\UAS.cPa also suppress the apoptosis observed in these clones.

The co-expression of SPARCScer\UAS.cPa suppresses the increased apoptosis observed in the posterior compartment of wandering third instar larval wing discs expressing wupAJF02172 under the control of Scer\GAL4en.PU.

The induction of somatic clones co-expressing l(2)glGD4047 and Ras85DV12.cUa.Scer\UAS under the control of Scer\GAL4Act.PU during second instar larval stage results in the accumulation of clonal cells in the wandering third instar larval fat body, which is enhanced by the additional co-expression of wupAJF02172; the induction of these clones also results in lethality before adulthood, which is partially rescued by the additional co-expression of wupAJF02172, although the surviving adults display a shorter life-span as compared to wild-type controls.

In wandering third instar larvae, somatic clones co-expressing l(2)glGD4047 and Ras85DV12.cUa.Scer\UAS under the control of Scer\GAL4Act.PU in both eye discs and wing discs exhibit a significant increase in the relative clonal area and hyperplasia as compared to control clones; wing disc clone cells localize both basally and apically in the disc. All of these phenotypes are suppressed by the additional co-expression of wupAJF02172, even leading to a significant decrease in the wing disc clonal area as compared to control clones.

Comments

The co-expression of wupAf06492 rescues the progressive decrease in the relative area of somatic clones expressing wupAJF02172 under the control of Scer\GAL4Act.PU in wandering third instar larval wing discs, even leading to a significant increase in the relative clonal area, as compared to control clones.
