UASp regulatory sequences are fused upstream of sequences encoding the Ada2b L isoform, including the Ada2b regulatory region up to position -400. The Ada2b coding sequence is tagged at the C-terminal end with EGFP. The protein can be expressed either under the control of its own regulatory sequences or, if a GAL4 driver is used, under the control of the UASp regulatory sequences.

Ada2bScer\UAS.P\T.T:Avic\GFP-EGFP is able to produce an 80% phenotypic rescue of Ada2bd842 mutants in the absence of a Scer\GAL4 driver. When expression of Ada2bScer\UAS.P\T.T:Avic\GFP-EGFP is driven by Scer\GAL4Act5C.PU the same degree of rescue of Ada2bd842 is seen as in the absence of the driver.
