UASt regulatory sequences drive expression of a mutant form of Hsap\APP that is fused to a signal peptide (Tag:SS(nec)) and carries F4D/E22G amino acid substitutions.
Female flies expressing Hsap\APPF4DE22G.Scer\UAS.T:SS-nec in the developing nervous system, under the control of Scer\GAL4elav-C155 are short lived as compared to controls. These flies exhibit more abundant protofibrils (HMW oligomers), which have been shown to be more toxic than fibrils, than in controls. Exposing these flies to 2, 20, and 200υM of the compound D737 from the embryonic stage onwards extends longevity. These flies also show a dramatic improvement in climbing ability.
