Expression of par-1HMS00405 under the control of Scer\GAL4ppk.PG does not affect the morphology or axonal projections of larval class IV dendritic arborizing neuron (c4da) in the ventral nerve cord but it leads to dendrite pruning defects in the pupal stage as no dendrite thinning is observed at 6h after puparium formation (APF) and the neurons retain dendrites attached to the cell body at 18h APF (by which point the pruning is completed in wild-type flies).

Embryos from mothers expressing par-1HMS00405 under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16 display enlarged cells with multiple centrosomes.

Expression of par-1HMS00405 under the control of Scer\GAL4SPARC-MI00329-GAL4 does not result in melanisation of the fat body.

Females heterozygous for par-1HMS00405 and MTD-Gal4 (consists of Scer\GAL4otu.T:Hsim\VP16, Scer\GAL4nos.PG and Scer\GAL4nos.UTR.T:Hsim\VP16) crossed to par-1HMS00405 males results in: "few eggs,47% hatching"; egg/embryonic phenotype: few eggs and tiny; maternal effect; oogenesis defect.

Expression of par-1HMS00405 under the control of either Scer\GAL4ey.PH or Scer\GAL4Bx-MS1096 reduces the size of the eye or wing respectively. When par-1HMS00405 is expressed under the control of Scer\GAL4hh-Gal4 the relative P-compartment area of the wing is reduced compared to controls. Expressing par-1HMS00405 under the control of Scer\GAL4GMR.PU results in increased eye disc apoptosis.

Eye disc clones expressing par-1HMS00405 under the control of Scer\GAL4unspecified are significantly smaller than the corresponding wild type controls.

The penetrance of the pruning defects observed in class IV dendritic arborizing neurons of pupae expressing par-1HMS00405 under the control of Scer\GAL4ppk.PG is significantly enhanced by co-expression of any of the following: tauScer\UAS.RA.T:Ivir\HA1, tauS184A.Scer\UAS.RA.T:Ivir\HA1, kat-60L1HMS00510 or shi1.Scer\UAS (the latter two on their own induce mild pruning defects too) or by combination with IKKεalice. The phenotype is weakly enhanced by co-expression of futschEP1419 and can be suppressed by combination with a single copy of tauMR22, while combination with one copy of any of the following: futschK68, EcR31, Rpn8k08003 fails to significantly modify the phenotype.

Maternal expression of par-1HMS00405 under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16 rescues the abnormal chromosome positioning of embryos from aPKCts/Df(2R)l4 mothers, but also results in enlarged cells with multiple centrosomes.

Expression of ykiScer\UAS.cZa suppresses the shrunken P-compartment phenotype seen in wing discs when par-1HMS00405 is expressed under the control of Scer\GAL4hh-Gal4.

Expression of par-1HMS00405 suppresses the increased P-compartment phenotype seen in wing discs when exGD12782 is expressed under the control of Scer\GAL4hh-Gal4.

Expression of par-1HMS00405 suppresses the wing overgrowth seen when exGD12782 is expressed under the control of Scer\GAL4Bx-MS1096.

Expression of par-1HMS00405 suppresses the overgrowth phenotype seen in wings expressing ftGD881 under the control of Scer\GAL4Bx-MS1096.

wtsx1 rescues the reduction in eye disc clone size seen when par-1HMS00405 is expressed under the control of Scer\GAL4unspecified, with the resulting clones similar in size to those seen with wtsx1 alone.
