Optogenetic activation (using Cnoc\ChR1::Csub\ChRCsChrimson.20xUAS.FRT.Venus with Scer\FLP1L.8xlexAop and 627 nm (red) light) of cells expressing both Scer\GAL4GMR18H11 and Ecol\lexAGMR67F03, in adults significantly increases sleep, compared to controls.

Optogenetic inhibition (using Gthe\ACR1UAS.d.EYFP with 540 nm (green) light) of Scer\GAL4GMR18H11 cells significantly reduces sleep, compared to controls.

Activation of Scer\GAL4GMR18H11-expressing neurons (using heat activation of a TrpA1 construct) significantly suppresses sleep, with the effect being strongest in the hours before and after dawn. Since this effect can be suppressed by inhibiting Scer\GAL4 expression in clock neurons (using Scer\GAL80) and Scer\GAL4GMR18H11 is normally active in a restricted subset of four to six Dh31-expressing DN1 clock neurons, this indicates that Dh31-expressing DN1 circadian clock neurons are intrinsically wake promoting.
