Scer\UAS regulatory and P\T promoter sequences drive expression of a cnn coding sequence which lacks the amino terminal 17 amino acids of the maternal cnn-RA transcript and in which 13 core amino acid residues in the conserved "motif 1" have been deleted. The cnn sequences are tagged with T:Avic\GFP[EGFP].

Embryos expressing cnn[Δ1.Scer\UAS.T:Avic\GFP-EGFP] under the control of Scer\GAL4[nos.UTR.T:Hsim\VP16] do not hatch. Centrosome duplication appears normal in these embryos, but centrosome separation fails. The mutant embryos also have reduced numbers of centrosomal satellites. Expression of cnn[Δ1.Scer\UAS.T:Avic\GFP-EGFP] under the control of Scer\GAL4[tub.PU] from late embryogenesis onwards is not zygotically lethal.
