The expression of bratHMS01121 under the control of Scer\GAL4slbo.2.6 does not lead to defects in border cell migration or cluster integrity.

Third instar larval brain tissue expressing bratHMS01121 under the control of Scer\GAL4da.PU exhibits overgrowth and no significant difference in cell death levels as compared to controls, and when this brain tissue (or that expressing bratHMS01121 under the control of Scer\GAL4Act5C.PI) is transplanted into the adult abdomen of a wild type host fly it exhibits strong tumorigenic growth, high levels of reactive oxygen species, metastasis, and a high proportion of defects in anaphase, as compared to control transplanted tissue.

The larval brain overgrowth exhibited upon expression of bratHMS01121 under the control of Scer\GAL4da.PU is suppressed when ZwKK108898, bskGD10555 or PepckGD9451 is co-expressed, and partially suppressed when Mfe2KK101732 or WwoxKK108459 is co-expressed. Of these co-expressed alleles, none induce increased cell death levels in the larval brain tissue except for co-expression of ZwKK108898, which leads to a significant increase in cell death. The proportion of defective anaphases seen in larval brains expressing bratHMS01121 under the control of Scer\GAL4da.PU is significantly elevated upon co-expression of bskGD10555 or Mfe2KK101732, but is not significantly changed upon co-expression of ZwKK108898, PepckGD9451 or WwoxKK108459. The tumor growth rate of larval brain explants expressing bratHMS01121 under the control of Scer\GAL4da.PU is suppressed upon co-expression of PepckGD9451 or Mfe2KK101732, but not ZwKK108898, bskGD10555 or WwoxKK108459.

The tumor growth rate of larval brain explants expressing bratHMS01121 under the control of Scer\GAL4Act5C.PI is suppressed upon co-expression of GpdhGD14102, Mdh1GD11625, or MenGL01234.
