Adulthood-only expression of Atg16HMS01347 under the control of Scer\GAL4esg.PU leads to a significant increase in the number of mitotic cells (phospho-H3 staining) in the adult midgut, as compared to controls. (The temporal regulation of expression is dependent on Gal80[ts] and induced by temperature shift to 29[o]C for 5-6 days during adulthood).

The expression of Atg16HMS01347 under the control of Scer\GAL4Cg.PA blocks the starvation-induced accumulation of acidic vesicles (LysoTracker-positive) in the early third instar larval fat body, as compared to controls. Under standard conditions, there is no change in the number of autophagosome-like structures (Atg8a-positive), as compared to controls.

The expression of Atg16HMS01347 under the control of Scer\GAL4pros-V1 leads to a significant increase in the number of pre-enteroendocrine cells, as compared to controls.

Expression of Atg16HMS01347 under the control of Scer\GAL4Myo31DF-NP0001 results in significantly delayed midgut degradation after pupariation, with increased size of the midgut and gastric caeca 0 hrs and 4 hrs after puparium formation, but no significant difference in autophagy levels 2 hrs after puparium formation, as compared to controls.

Expression of Atg16HMS01347 under the control of Scer\GAL4cg.PU results in a decrease in starvation-induced autophagy in the larval fat body, as compared to controls.

Expression of Atg16HMS01347 under the control of Scer\GAL4Mef2.PR is able to significantly suppress the formation of autophagosomes in the muscles of chloroquine-treated larvae starved on low-nutrient food for 6 hours.

Expression of Atg16HMS01347 using Scer\GAL4Act5C.PP results in basal autophagy defects in larval fat body cell somatic clones.
