Expression of CdsANIG.7962R in both the larval fat body and salivary gland, under the control of Scer\GAL4ppl.PP, causes an ectopic lipid droplet phenotype.

Expression of CdsANIG.7962R under the control of Scer\GAL4αTub84B.PL, results in these larvae accumulating excess fat in many non-adipose tissues, including the salivary gland, proventriculus, hind gut, Malpighian tubules, and trachea. Compared to controls, Scer\GAL4αTub84B.PL-->CdsANIG.7962R animals exhibit a much higher level of triacylglycerol.

Expression of CdsANIG.7962R under the control of Scer\GAL4αTub84B.PL results in a significant reduction in organ size, compared to controls. The imaginal discs, salivary glands, and brain are smaller in these flies, compared to controls. While cell number is not changed in these mutants, cell size is greatly reduced.

The levels of diacylglycerol and phosphatidylethanolamine are significantly increased in fat body cells expressing CdsANIG.7962R under the control of Scer\GAL4αTub84B.PL.

Expression of CdsANIG.7962R using Scer\GAL4ppl.PP or Scer\GAL4elav.PU, but not Scer\GAL4Sgs3.PD, results in ectopic lipid storage in the salivary gland.

Overexpression of Akt1Scer\UAS.Exel or constitutively active Pi3K92EScer\UAS.T:Hsap\MYC,T:Hsap\CAAX in the salivary glands, under the control of Scer\GAL4ppl.PP, fully suppresses the small cell size phenotype found upon knockdown of CdsA through expression of CdsANIG.7962R. The ectopic lipid accumulation phenotype of CdsANIG.7962R mutants is partially rescued by overexpression of Pi3K92EScer\UAS.T:Hsap\MYC,T:Hsap\CAAX but not Akt1Scer\UAS.Exel.

A Df(3R)Pi3K92EA or Akt104226 heterozygous background results in a reduction in the size of fat cells by around 50% in flies expressing CdsANIG.7962R under the control of Scer\GAL4ppl.PP.

CdsANIG.7962R and bbcGD1980 double mutants exhibit a synergistic effect, resulting in a drastic decrease in fat cell size. Salivary gland cell size is unaffected.
