A 3.0kb full length vav coding sequence is expressed under the control of Scer\UAS regulatory sequences and tagged at the C-terminal with T:Ivir\HA1. Two alterations to the sequence are present, causing M53T and a silent codon change in codon 313.

Embryos expressing vav[Scer\UAS.T:Ivir\HA1] in the epidermis using the Scer\GAL4[69B] driver are indistinguishable from wild type. Embryos expressing vav[Scer\UAS.T:Ivir\HA1] in the mesoderm using the Scer\GAL4[how-24B] driver do not display any obvious muscle defects. Embryos expressing vav[Scer\UAS.T:Ivir\HA1] under the control of the nervous system driver Scer\GAL4[elav.PLu] are indistinguishable from wild type. Caudal visceral cell (CVM) migration is comparable to wild type when vav[Scer\UAS.T:Ivir\HA1] is expressed in embryos under the control of the Scer\GAL4[447.2] driver. Expression of vav[Scer\UAS.T:Ivir\HA1] under the control of the tracheal system driver Scer\GAL4[btl.PS] has no effect on tracheal development.

Expression of vav[Scer\UAS.T:Ivir\HA1] under the control of either the Scer\GAL4[arm.PS] or the Scer\GAL4[αTub84B.PL] driver rescues the semi-lethality seen in vav[2] male homozygotes. Expression of vav[Scer\UAS.T:Ivir\HA1] under the control of either the Scer\GAL4[arm.PS] or the Scer\GAL4[αTub84B.PL] driver rescues the semi-lethality seen in vav[3] male homozygotes. Expression of vav[Scer\UAS.T:Ivir\HA1] under the control of Scer\GAL4[arm.PS] partially rescues the midline crossing phenotype seen in vav[2] homozygotes. Expression of vav[Scer\UAS.T:Ivir\HA1] under the control of Scer\GAL4[arm.PS] partially rescues the midline crossing phenotype seen in vav[3] homozygotes. Expression of vav[Scer\UAS.T:Ivir\HA1] under the control of Scer\GAL4[GMR.PU] fails to rescue the photoreceptor targeting phenotype seen in vav[1] homozygotes.
