A constitutively active form of Vav, comprising a deletion of the N-terminus (amino acids 1-207), is expressed under the control of UASt regulatory sequences and tagged at the C-terminal end with Tag:HA. A silent codon change is present in codon 313.

Expression of constitutively active vavΔ1-207.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PU results in severe disruption to adult eye morphology.

Expression of vavΔ1-207.Scer\UAS.T:Ivir\HA1 in the epidermis of embryos from stage 9 onwards using the Scer\GAL469B driver results in dorsal closure failure. The lateral epidermal cells are disorganised and fail to elongate.

Expression of vavΔ1-207.Scer\UAS.T:Ivir\HA1 in the embryonic mesoderm using the Scer\GAL4how-24B driver results in inhibition of myoblast fusion.

Expression of vavΔ1-207.Scer\UAS.T:Ivir\HA1 under the control of the nervous system driver Scer\GAL4elav.PLu results in embryos that display axon guidance defects such as misrouting of longitudinal axons across the midline and gaps between segments. The axons appear thicker than in wild type embryos. Axons also fail to extend between the dorsal and lateral clusters of the peripheral nervous system.

When vavΔ1-207.Scer\UAS.T:Ivir\HA1 is expressed in the caudal visceral mesoderm (CVM) cells under the control of Scer\GAL4447.2 they fail to migrate towards the anterior pole of the embryo. The CVM cells remain viable at the posterior end of the embryo, indicating that cell survival is unaffected.

Expression of vavΔ1-207.Scer\UAS.T:Ivir\HA1 under the control of the tracheal system driver Scer\GAL4btl.PS results in a variety of defects in the embryo including severe truncation of the tracheal dorsal trunk, misguided dorsal branching and limited terminal branching differentiation.
