Expression of Pk92B[ΔN.Scer\UAS.T:Zzzz\FLAG] under the control of Scer\GAL4[pnr-MD237] results in an increase in melanisation of the adult thorax cuticle in the domain of Scer\GAL4[pnr-MD237] expression.
Flies expressing Pk92B[ΔN.Scer\UAS.T:Zzzz\FLAG] under the control of Scer\GAL4[pnr-MD237] exhibit black pigmentation in the thorax cuticle. Ubiquitous expression of Pk92B[ΔN.Scer\UAS.T:Zzzz\FLAG] using Scer\GAL4[hs.PU], induced by a 1 hour heat shock during the pupal stage, results in black pigmentation only in the lower medial region of the thorax cuticle, together with a defect in wing expansion.

Co-expression of slim[dsRNA.Scer\UAS] has no effect on the increase in melanisation of the adult thorax cuticle seen in flies expressing Pk92B[ΔN.Scer\UAS.T:Zzzz\FLAG] under the control of Scer\GAL4[pnr-MD237].
Co-expression of Mpk2[DN.Scer\UAS], Mpk2[NIG.5475R], p38b[NIG.7393R], or lic[GD7546] abolishes the thorax pigmentation defects seen in Pk92B[ΔN.Scer\UAS.T:Zzzz\FLAG], Scer\GAL4[pnr-MD237] flies. Co-expression of p38c[KK103439] has only a marginal suppressive effect. Co-expression of Mkk4[NIG.9738R] attenuates the thorax pigmentation defects seen in Pk92B[ΔN.Scer\UAS.T:Zzzz\FLAG], Scer\GAL4[pnr-MD237] flies. Co-expression of hep[NIG.4153R] or hemizygosity for hep[1] does not suppress the thorax pigmentation defects seen in Pk92B[ΔN.Scer\UAS.T:Zzzz\FLAG], Scer\GAL4[pnr-MD237] flies. Co-expression of e[Scer\UAS.cWa] abolishes the thorax pigmentation defects seen in Pk92B[ΔN.Scer\UAS.T:Zzzz\FLAG], Scer\GAL4[pnr-MD237] flies. In a y[1] hemizygous background, pigmentation in the thorax of Pk92B[ΔN.Scer\UAS.T:Zzzz\FLAG], Scer\GAL4[pnr-MD237] flies is brown rather than black, particularly in the scutellum. Co-expression of either ple[dsRNA.Scer\UAS] or Ddc[NIG.10697R] abolishes the thorax pigmentation defects seen in Pk92B[ΔN.Scer\UAS.T:Zzzz\FLAG], Scer\GAL4[pnr-MD237] flies. Co-expression of Hr38[dsRNA.Scer\UAS] reduces the thorax pigmentation defects seen in Pk92B[ΔN.Scer\UAS.T:Zzzz\FLAG], Scer\GAL4[pnr-MD237] flies at 18[o]C.
