Expression of DNApol-ε[NIG.6768R] under the control of either Scer\GAL4[GMR.PS], Scer\GAL4[en-e16E], Scer\GAL4[sd.PU] or Scer\GAL4[Bx-MS1096] does not result in a detectable phenotype. Expression of DNApol-ε[NIG.6768R] under the control of Scer\GAL4[Act5C.PU] results in a reduction in size of the salivary gland, brain lobes and imaginal discs compared to wild type. Expression of DNApol-ε[NIG.6768R] under the control of Scer\GAL4[ey.PU] results in a small eye, with a reduction in the number of ommatidia. Incorporation of BrdU in the S phase zone posterior to the morphogenetic furrow is significantly reduced in eye imaginal discs in the animals. Expression of DNApol-ε[NIG.6768R] under the control of Scer\GAL4[ap-md544] results in an outstretched wing phenotype. Expression of DNApol-ε[NIG.6768R] under the control of Scer\GAL4[l(1)BP-1] results in a small salivary gland. The number and size of the salivary gland cell nuclei are decreased relative to wild type. The incorporation of BrdU in larvae 48 hours after hatching is severely decreased compared to wild type.
Expression of DNApol-ε[NIG.6768R] under the control of Scer\GAL4[Act5C.PI] results in approximately 50% larval and pupal lethality. Larvae expressing DNApol-ε[NIG.6768R] under the control of Scer\GAL4[Act5C.PI] show increased sensitivity to DNA damage in response to bleomycin compared to wild type.

Co-expression of DNApol-ε[Ct.Scer\UAS.T:Ivir\HA1] rescues the small eye phenotype and mitotic cycle defects seen in the eye imaginal disc which are caused by expression of DNApol-ε[NIG.6768R] under the control of Scer\GAL4[ey.PU]. Co-expression of DNApol-ε[Ct.Scer\UAS.T:Ivir\HA1] does not rescue the endocycle defects seen in the salivary glands of animals expressing DNApol-ε[NIG.6768R] under the control of Scer\GAL4[l(1)BP-1].
