Adulthood-only expression of upd1UAS.cUa under the control of Scer\GAL4NP5130 (and GAL80[ts] for the temporal control of expression) induces an increase in the number of progenitor cells in the midgut, as well as an increase in mitotic index.

Expression of upd1UAS.cUa under the control of Scer\GAL4rn-GAL4-5 (with tub-Gal80[ts] restricting expression to third instar larvae) does not lead to ectopic wing pouches in wing discs or ectopic wings when compared to controls.

Expression of upd1UAS.cUa under the control of Scer\GAL4GMR15B03 or Scer\GAL4dpp.blk1 leads to disruption of thorax and loss of many mechanosensory chaetas when compared to control adults.

Adult flies expressing upd1Scer\UAS.cUa driven by Scer\GAL4esg-NP7397 combined with tub-Gal80[ts] (for temporal control) display significant increase in midgut stem cell proliferation (assessed by number of pH3-positive cells).

The expression of upd1Scer\UAS.cUa under the control of Scer\GAL4ptc.PU leads to a significant decrease in the size of mesonotum primordia size, from which ectopic wing primordium structures frequently emerge, as compared to controls.

Expression of upd1Scer\UAS.cUa under the control of Scer\GAL4esg-NP5130 (in combination with a Gal80[ts] transgene to restrict expression to the adult stage) results in a significant increase in the number of mitotic cells in the midgut, as compared to controls.

Scer\GAL4ptc-559.1-mediated expression of upd1Scer\UAS.cUa results in overgrown wing discs.

Expression of upd1Scer\UAS.cUa under the control of Scer\GAL4tim.PE results in arrhythmic locomotor activity under constant darkness conditions.

Expression of upd1Scer\UAS.cUa under the control of Scer\GAL4elav.Switch.PO in the presence of RU486 results in defects in locomotor activity rhythms (only 33.3% of flies show rhythmic behaviour under constant darkness conditions).

Expression of upd1Scer\UAS.cUa under the control of Scer\GAL4esg-NP7397 and Scer\GAL80ts.αTub84B induces intestinal stem cell proliferation.

No tumors are seen when upd1Scer\UAS.cUa is expressed in eye-antennal clones under the control of Scer\GAL4Act.PU. Inhibition of apoptosis in upd1Scer\UAS.cUa-expressing cells does not induce tumors.

Wing discs expressing upd1Scer\UAS.cUa under the control of Scer\GAL4Bx-MS1096 show an expansion of the disc epithelium and increased epithelial folding.

Co-expression of upd1UAS.cUa and egrUAS.cUa under the control of Scer\GAL4rn-GAL4-5 (with tub-Gal80[ts] restricting expression to third instar larvae) leads to ectopic wings compared to controls.

Expression of upd1UAS.cUa under the control of Scer\GAL4rn-GAL4-5 (with tub-Gal80[ts] restricting expression to third instar larvae) in CtBP87De-10/+ background does not lead to ectopic wing pouches in wing discs or ectopic wings when compared to controls.

The increased number of intestinal progenitor cells in mitosis characteristic for adult flies expressing upd1Scer\UAS.cUa driven by Scer\GAL4esg-NP7397 (combined with tub-Gal80[ts] for temporal control) is not suppressed by co-expression of RetGD45.

Co-expression of bunGD1392 suppresses the increased number of mitotic cells seen in the midguts of flies expressing upd1Scer\UAS.cUa under the control of Scer\GAL4esg-NP5130.

Intestinal stem cell proliferation found in flies expressing upd1Scer\UAS.cUa under the control of Scer\GAL4esg-NP7397 and Scer\GAL80ts.αTub84B is completely inhibited when Egfr or Ras85D is down-regulated (through expression of either EgfrdsRNA.Scer\UAS.cUa or Ras85DdsRNA.Scer\UAS.cUa under the control of Scer\GAL4esg-NP7397 and Scer\GAL80ts.αTub84B).

Intestinal stem cell proliferation found in flies expressing upd1Scer\UAS.cUa under the control of Scer\GAL4esg-NP7397 and Scer\GAL80ts.αTub84B is completely inhibited in Ras85DΔC40B mutant clones.

Co-expression of upd1Scer\UAS.cUa and spis.Scer\UAS in intestinal stem cells, under the control of Scer\GAL4Myo31DF-NP0001 and Scer\GAL80ts.αTub84B induces a much higher intestinal stem cell mitotic index than that induced by the activation of either transgene alone.

No tumors are seen when upd1Scer\UAS.cUa is expressed under the control of Scer\GAL4Act.PU in scrib1 mutant eye-antennal clones.

No tumors are seen when upd1Scer\UAS.cUa is expressed under the control of Scer\GAL4Act.PU in wild type eye-antennal disc cells that are adjacent to scrib1 mutant cells.

Eye-antennal disc clones that simultaneously express Ras85DV12.Scer\UAS and upd1Scer\UAS.cUa under the control of Scer\GAL4Act.PU grow into large invasive tumors.

Eye-antennal disc clones that simultaneously express Ras85DV12.Scer\UAS, upd1Scer\UAS.cUa and upd2Scer\UAS.cUa under the control of Scer\GAL4Act.PU grow into large invasive tumors that are larger than those seen when Ras85DV12.Scer\UAS is co-expressed with either upd1Scer\UAS.cUa or upd2Scer\UAS.cUa alone.

Expression of bskDN.Scer\UAS does not suppress the tumors seen in eye-antennal disc clones that simultaneously express Ras85DV12.Scer\UAS and upd1Scer\UAS.cUa under the control of Scer\GAL4Act.PU.

Expression of domeΔCYT.Scer\UAS in eye-antennal disc clones that express Ras85DV12.Scer\UAS and upd1Scer\UAS.cUa under the control of Scer\GAL4Act.PU suppresses the overgrowth and invasion of the nerve cord.

Comments

The increased attraction to food cues, increased food consumption and higher body weight of adult flies expressing upd1JF03149 under the control of Scer\GAL4elav.PU can be rescued by expression of upd1Scer\UAS.cUa.
