Scer\UAS regulatory sequences drive expression of the "Yellow Cameleon 2.0" Ca[2+] sensor. This protein consists of two different chromophores (ECFP and EYFP variants of Avic\GFP respectively) separated by a calcium binding region (T:Xlae\Cam). Ca[2+]-binding to T:Xlae\Cam results in a conformational change that can be detected as a change in the efficacy of fluorescence resonance energy transfer (FRET) between the two Avic\GFP variants.

Scer\GAL4[elav-C155]-mediated expression of Avic\GFP[Cameleon2.0.Scer\UAS] does not affect the neurological phenotype of GluRIIA[AD9] mutants. Scer\GAL4[elav-PG159]-mediated expression of Avic\GFP[Cameleon2.0.Scer\UAS] does not affect the phenotype of pAbp[k10109]/+ mutant larvae: these mutants still have larger evoked postsynaptic currents and significantly more boutons compared to wild type.
