Expression of rigNIG.11171R under the control of Scer\GAL4Mef2.PR has no effect on flight ability of adult flies, mobility of the third instar larvae or shape of pupae and Scer\GAL4elav-C155-driven expression does not impair adult viability.

Expression of rigNIG.11171R driven by Scer\GAL4Mef2.PR (without UAS-dicer2 to enhance RNAi efficiency) does not result in flight or lifespan defects.

Flies co-expressing two different insertions of rigNIG.11171R (P{NIG.11171R}1 and P{NIG.11171R}2) ubiquitously under the control of Scer\GAL41032.hx, Scer\GAL4da.PU or Scer\GAL4Act5C.PU are completely lethal. When expressed in muscle lineages under the control of Scer\GAL4Mef2.PR viable flies are seen at both 25[o]C and 29[o]C. When Dcr-2Scer\UAS.cDa is also co-expressed almost complete lethality is seen at 29[o]C. Flies co-expressing Dcr-2Scer\UAS.cDa at 25[o]C show a progressive decline in flight ability. Complete lethality is seen at both 25[o]C and 29[o]C when rigNIG.11171R is expressed under the control of two further muscle drivers, Scer\GAL4c179 or Scer\GAL4how-24B. Expression of rigNIG.11171R in the somatic musculature under the control of Scer\GAL4G7 results in viability at 25[o]C but semi-lethality at 29[o]C. Pan-neuronal expression under the control of Scer\GAL4elav.PU results in viability at 25[o]C, both in the presence or absence of Dcr-2Scer\UAS.cDa. Flies are viable at 29[o]C in the absence of Dcr-2Scer\UAS.cDa, but complete lethality is seen when it is present. Flies co-expressing Dcr-2Scer\UAS.cDa at 25[o]C show flight and climbing defects compared to controls. Some flies have curved, rather than straight wing blades and in some cases the wings are crumpled and unopened. Expression in motor neurons under the control of Scer\GAL4RapGAP1-OK6 is viable at 25[o]C, in the absence of Dcr-2Scer\UAS.cDa, and semi-viable flies when it is present. Flies are semi-lethal at 29[o]C in the absence of Dcr-2Scer\UAS.cDa, but completely lethal when it is present.
