Inhibition (using Ctet\TeTxLCFRT.stop.UAS or Hsap\KCNJ2FRT.STOP.10x.UAS.GFP) of Scer\GAL4dsx.Δ2 neurons expressing Scer\FLP1L.8xlexAop under the control of Ecol\lexAGMR13H01 in males results in almost no sine song during courtship, but a similar amount of pulse song to controls; the pulse song has an increase in inter-pulse interval, which is significant in the case of Ctet\TeTxLCFRT.stop.UAS, but no difference in carrier frequency or amplitude, compared to controls.

Acute optogenetic activation (using Cnoc\ChR1Cs.20xUAS.FRT.Venus with 660 nm (red) light activation) of Scer\GAL4dsx.Δ2 neurons expressing Scer\FLP1L.8xlexAop under the control of Ecol\lexAGMR13H01 in males, in the absence of females, results in single wing extension and sine song similar to that of controls courting a female, but little pulse song.

Thermogenetic activation (using TrpA1FRT.stop.UAS.Tag:MYC at 29[o]C with Scer\FLP1L.8xlexAop) of cells expressing Scer\GAL4dsx.Δ2 and Ecol\lexAGMR71G01 in adult males significantly increases aggressive lunging behavior and wing extensions towards other males, compared to controls.
