Flies expressing either the P{UAS-Irk2.DN}5.1 or P{UAS-Irk2.DN}5.2 insertion lines of P{UAS-Irk2.DN} are lethal when crossed to Scer\GAL4Act5C.PI. The P{UAS-Irk2.DN}13 insertion line survives to adulthood when raised at 25[o]C, although only 20% survive when flies are raised at 18[o]C.

Only 17% of larvae expressing Irk2DN.Scer\UAS (insertion line P{UAS-Irk2.DN}5.1) under the control of Scer\GAL4Act5C.PI are able to hatch. No larvae survive beyond 48 hours.

All of the female flies resulting from the expression of P{UAS-Irk2.DN} (insertion line P{UAS-Irk2.DN}5.1 in the wing disc under the control of Scer\GAL4Bx-MS1096 at 25[o]C display wing defects. 6% of wings display moderate defects, which include wings that are small, collapsed or blistered, as well as those showing branching of the posterior cross-vein, L2-3 collapse, L4 bifurcation, incomplete L5 vein and wing bristle transformations. 94% of flies showed more severe defects, including partial or complete loss of the wing blade. For the P{UAS-Irk2.DN}5.2 insertion line, 42% show moderate wing defects and only 4% have severe defects. The phenotypes are more severe in males, and when flies are raised at lower temperatures.

Expression of P{UAS-Irk2.DN} (insertion line P{UAS-Irk2.DN}5.2) under the control of Scer\GAL4en-e16E causes wing vein and blistering defects.

Expression of Irk2DN.Scer\UAS (insertion line P{UAS-Irk2.DN}5.1) under the control of either Scer\GAL4en-e16E or Scer\GAL4A9 results in a reduction in wing disc size. Less than 5% of either genotype survives beyond the early larval stage. Expression of the P{UAS-Irk2.DN}13 under the control of Scer\GAL4A9 insertion line also results in a reduction in disc size.

Wing discs expressing Irk2DN.Scer\UAS (P{UAS-Irk2.DN}5.1) under the control of Scer\GAL4Bx-MS1096 are fragile and thinned in the wing pouch region. Apoptosis (TUNEL staining) occurs in the wing pouch area.

One copy of dpphr92 enhances the wing defects seen when Irk2DN.Scer\UAS is expressed under the control of Scer\GAL4Bx-MS1096. 84% of wings from female flies expressing the P{UAS-Irk2.DN}5.2 insertion line display defects, with 29% of these considered to be severe. When the stronger P{UAS-Irk2.DN}5.1 insertion line is expressed 97% of wings from female flies display severe defects.

One copy of tkv7 enhances the wing defects seen when Irk2DN.Scer\UAS is expressed under the control of Scer\GAL4Bx-MS1096. Total wing defects in females expressing the P{UAS-Irk2.DN}5.2 insertion line are increased from 46% to 72% of wings. Total wing defects in females expressing the stronger P{UAS-Irk2.DN}5.1 line are increased from 94% to 99% of wings.

Expression of BacA\p35Scer\UAS.cHa suppresses the apoptosis seen in wing discs expressing Irk2DN.Scer\UAS under the control of Scer\GAL4Bx-MS1096, although apoptosis is still seen in the periphery of the wing outside of the region of Scer\GAL4Bx-MS1096 expression. The severe wing phenotypes are not suppressed.
