Expressing shotGL01286 under the control of Scer\GAL4SPARC-MI00329-GAL4 results in third instar larval adipocytes presenting extracellular matrix accumulations extending to the cell body (Vkg.GFP).

Expressing shotGL01286 under the control of Scer\GAL4Cg.PA results in third instar larval adipocytes presenting enlarged, deformed and highly eccentric nuclei, aberrant matrix accumulations (Trol.GFP) extending to the cell body, and plasma membrane invaginations that concentrates actin filaments, myosin and Rho1. In agreement, mitochondria, endoplasmic reticulum exit sites and endocytic or recycling vesicles lose their homogeneous distribution throughout the cytoplasm and seem to gravitate toward a single focus near the nucleus. These adipocytes also exhibit a large juxtanuclear focus of microtubules, including non-centrosomal minus-ends (Patronin.GFP), in contrast to a normal perinuclear distribution in controls.

Expressing shotGL01286 under the control of Scer\GAL4NP0001 and Scer\GAL4Ugt36A1.PK causes aberrant juxtanuclear microtubule foci and mispositioned nuclei in the midgut enteroblasts and garland filter cells, respectively.

Expression of shotGL01286 under the control of Scer\GAL4elav.PU results in overgrowth (increase in bouton number) at the neuromuscular junction (NMJ) in third instar larvae, while expression under the control of Scer\GAL4how-24B has no effect on bouton number at the NMJ.

Axons expressing shotGL01286 under the control of Scer\GAL4tey-5053A show enhanced regenerative axonal sprouting compared to controls after a crushing injury.

The enhanced regenerative axonal sprouting seen in axons expressing shotGL01286 under the control of Scer\GAL4tey-5053A compared to controls after a crushing injury is eliminated if they are co-expressing wndVDRC.cUa.
