Maternal expression of cystHMS01750 under the control of Scer\GAL4VP16.mat.αTub67C leads to embryonic lethality, associated with epithelial/cuticular defects, namely many small epithelial vesicles or cysts and small cuticle shields, instead of the large continuous epithelial sheet observed in controls. This cuticle phenotype is enhanced by maternal heterozygosity for Df(2L)BSC301.

The embryos show progressive adherens junction fragmentation during gastrulation ( DEca::GFP ) so that many cells, particularly in the ventral ectoderm, lose the Adherens Junctions; other cells show focal concentration of Adherens Junction components, suggesting the beginning of cyst formation. The epithelial cysts are first seen at the end of gastrulation and can be followed throughout the rest of development; these cysts show an apparently normal polarized distribution of Crb, aPKc, Arm and Yrt proteins.

While the maternal expression of cystHMS01750 under the control of Scer\GAL4VP16.mat.αTub67C does not greatly affect the planar polarization of Baz protein in embryos, additional maternal heterozygosity for Df(2L)BSC301 leads to Baz hyper-polarization.

Maternal co-expression of cystHMS01750 and scribHMS01490 under the control of Scer\GAL4VP16.mat.αTub67C leads to embryonic cuticle defects that are similar to those induced by the expression of scribHMS01490 alone (i.e. epidermal cell clusters surrounded by cuticle), and milder than those induced by the expression of cystHMS01750 alone.
